Sullivan,Jaakko Tuomilehto,Ming T. Tsuang,Hugh Watkins,Hugh Watkins,James G. Wilson,Mark J. Daly,Mark J. Daly,Daniel G. MacArthur,Daniel G. MacArthur +106 more
TL;DR: The aggregation and analysis of high-quality exome (protein-coding region) DNA sequence data for 60,706 individuals of diverse ancestries generated as part of the Exome Aggregation Consortium (ExAC) provides direct evidence for the presence of widespread mutational recurrence.
Journal ArticleDOI
Clonal Hematopoiesis and Blood-Cancer Risk Inferred from Blood DNA Sequence
Giulio Genovese,Anna K. Kähler,Robert E. Handsaker,Johan Lindberg,Samuel A. Rose,Samuel F. Bakhoum,Kimberly Chambert,Eran Mick,Benjamin M. Neale,Menachem Fromer,Shaun Purcell,Oscar Svantesson,Mikael Landén,Martin Höglund,Sören Lehmann,Stacey Gabriel,Jennifer L. Moran,Eric S. Lander,Patrick F. Sullivan,Pamela Sklar,Henrik Grönberg,Christina M. Hultman,Steven A. McCarroll +22 more
TL;DR: Clonal hematopoiesis with somatic mutations is readily detected by means of DNA sequencing, is increasingly common as people age, and is associated with increased risks of hematologic cancer and death.
Journal ArticleDOI
Schizophrenia risk from complex variation of complement component 4
Aswin Sekar,Aswin Sekar,Allison R. Bialas,Heather de Rivera,Heather de Rivera,Avery Davis,Avery Davis,Timothy R. Hammond,Nolan Kamitaki,Nolan Kamitaki,Katherine Tooley,Katherine Tooley,Jessy Presumey,Matthew A. Baum,Vanessa Van Doren,Giulio Genovese,Giulio Genovese,Samuel A. Rose,Robert E. Handsaker,Robert E. Handsaker,Mark J. Daly,Mark J. Daly,Michael C. Carroll,Beth Stevens,Beth Stevens,Steven A. McCarroll,Steven A. McCarroll +26 more
TL;DR: It is found that many structurally diverse alleles of the complement component 4 (C4) genes generated widely varying levels of C4A and C4B expression in the brain, with each common C4 allele associating with schizophrenia in proportion to its tendency to generate greater expression of C 4A.
Journal ArticleDOI
De novo mutations in schizophrenia implicate synaptic networks
Menachem Fromer,Andrew Pocklington,David J. Kavanagh,Hywel Williams,Sarah Dwyer,Padhraig Gormley,Lyudmila Georgieva,Elliott Rees,Priit Palta,Douglas M. Ruderfer,Noa Carrera,Isla Humphreys,Jessica S. Johnson,Panos Roussos,Douglas Barker,Eric Banks,Vihra Milanova,Seth G. N. Grant,Eilis Hannon,Samuel A. Rose,Kimberly Chambert,Milind Mahajan,Edward M. Scolnick,Jennifer L. Moran,George Kirov,Aarno Palotie,Steven A. McCarroll,Peter Holmans,Pamela Sklar,Michael John Owen,Shaun Purcell,Michael Conlon O'Donovan +31 more
TL;DR: Genes affected by mutations in schizophrenia overlap those mutated in autism and intellectual disability, as do mutation-enriched synaptic pathways, and pathophysiology shared with other neurodevelopmental disorders.
Journal ArticleDOI
The cis-Regulatory Atlas of the Mouse Immune System
Hideyuki Yoshida,Caleb A. Lareau,Ricardo Ramirez,Samuel A. Rose,Barbara Maier,Aleksandra Wroblewska,Fiona Desland,Aleksey Chudnovskiy,Arthur Mortha,Claudia X. Dominguez,Julie Tellier,Edy Y. Brown,Sara Mostafavi,Jason D. Buenrostro,Jason D. Buenrostro,Christophe Benoist +27 more
TL;DR: This cis-regulatory atlas provides a trove of information on transcriptional regulation through immune differentiation and a foundational scaffold to define key regulatory events throughout the immunological genome.
