Abstract: Three crude oil bioremediation techniques were applied in a randomized block field experiment simulating a coastal oil spill. Four treatments (no oil control, oil alone, oil plus nutrients, and oil plus nutrients plus an indigenous inoculum) were applied. In situ microbial community structures were monitored by phospholipid fatty acid (PLFA) analysis and 16S rDNA PCR-denaturing gradient gel electrophoresis (DGGE) to (i) identify the bacterial community members responsible for the decontamination of the site and (ii) define an end point for the removal of the hydrocarbon substrate. The results of PLFA analysis demonstrated a community shift in all plots from primarily eukaryotic biomass to gram-negative bacterial biomass with time. PLFA profiles from the oiled plots suggested increased gram-negative biomass and adaptation to metabolic stress compared to unoiled controls. DGGE analysis of untreated control plots revealed a simple, dynamic dominant population structure throughout the experiment. This banding pattern disappeared in all oiled plots, indicating that the structure and diversity of the dominant bacterial community changed substantially. No consistent differences were detected between nutrient-amended and indigenous inoculum-treated plots, but both differed from the oil-only plots. Prominent bands were excised for sequence analysis and indicated that oil treatment encouraged the growth of gram-negative microorganisms within the α-proteobacteria and Flexibacter-Cytophaga-Bacteroides phylum. α-Proteobacteria were never detected in unoiled controls. PLFA analysis indicated that by week 14 the microbial community structures of the oiled plots were becoming similar to those of the unoiled controls from the same time point, but DGGE analysis suggested that major differences in the bacterial communities remained.

Abstract: Basic ionic liquids (ILs) based on a phosphonium hydroxide derivative can be tuned for CO{sub 2} capture by varying the weak proton donors, which have different pK{sub a} values. The stability, absorption capacity, and absorption enthalpy of the ILs could be easily tuned: the best IL for CO{sub 2} capture has good stability (>300 C), energy saving (ca. 56 kJ mol{sup -1}), and equimolar absorption capability.

Abstract: An integrated system for rapid PCR-based analysis on a microchip has been demonstrated. The system couples a compact thermal cycling assembly based on dual Peltier thermoelectric elements with a microchip gel electrophoresis platform. This configuration allows fast (∼1 min/cycle) and efficient DNA amplification on-chip followed by electrophoretic sizing and detection on the same chip. An on-chip DNA concentration technique has been incorporated into the system to further reduce analysis time by decreasing the number of thermal cycles required. The concentration injection scheme enables detection of PCR products after performing as few as 10 thermal cycles, with a total analysis time of less than 20 min. The starting template copy number was less than 15 per injection volume.

Abstract: A variety of ecological questions now require the study of large regions and the understanding of spatial heterogeneity. Methods for spatial-temporal analyses are becoming increasingly important for ecological studies. A grid cell based spatial analysis program (SPAN) is described and results of landscape pattern analysis using SPAN are presentedd. Several ecological topics in which geographic information systems (GIS) can play an important role (landscape pattern analysis, neutral models of pattern and process, and extrapolation across spatial scales) are reviewed. To study the relationship between observed landscape patterns and ecological processes, a neutral model approach is recommended. For example, the expected pattern (i.e., neutral model) of the spread of disturbance across a landscape can be generated and then tested using actual landscape data that are stored in a GIS. Observed spatial or temporal patterns in ecological data may also be influenced by scale. Creating a spatial data base frequently requires integrating data at different scales. Spatial is shown to influence landscape pattern analyses, but extrapolation of data across spatial scales may be possible if the grain and extent of the data are specified. The continued development and testing of new methods for spatial-temporal analysis will contribute to a general understanding of landscape dynamics.
