Binding activities of a repertoire of single immunoglobulin variable domains secreted from Escherichia coli

TLDR
Isolated variable domains may offer an alternative to monoclonal antibodies and serve as the key to building high-affinity human antibodies and the name 'single domain antibodies (dAbs)' is suggested for these antigen binding demands.
Abstract
IN antibodies, a heavy and a light chain variable domain, VH and VL, respectively, pack together and the hypervariable loops on each domain contribute to binding antigen1–4. We find, however, that isolated VH domains with good antigen-binding affinities can also be prepared. Using the polymerase chain reaction5, diverse libraries of VH genes were cloned from the spleen genomic DNA of mice immunized with either lysozyme or keyhole-limpet haemocyanin. From these libraries, VH domains were expressed and secreted from Escherichia coli. Binding activities were detected against both antigens, and two VH domains were characterized with affinities for lysozyme in the 20 nM range. Isolated variable domains may offer an alternative to monoclonal antibodies and serve as the key to building high-affinity human antibodies. We suggest the name 'single domain antibodies (dAbs)' for these antigen binding demands.

The effect of water activity on the association constant and the enthalpy of reaction between lysozyme and the specific antibodies D1.3 and D44.1

TL;DR: The reactions of lysozyme with the specific monoclonal antibody D1.3, its Fv fragment and a mutant of the Fv, were studied under conditions of reduced water activity through the addition of the cosolutes glycerol, ethanol, dioxane and methanol to determine enthalpy of reactions and affinity constants.

Folding and Assembly of an Antibody Fv Fragment, a Heterodimer Stabilized by Antigen

TL;DR: The analysis of the folding pathway of the domains VH and VL in isolation and in non-covalent and covalent assemblies should provide helpful insights into the folding of multimeric proteins in general, and for the further engineering of stable and well-folding antibody fragments in particular.

Phage Display Technology in Antibody Engineering: Design of Phagemid Vectors and in vitro Maturation Systems

TL;DR: The future will reveal if a combination of phage display technology with, e.g., in vitro immunization or primary/secondary immunization of Scid mice, transplanted with a human immune system (Carlsson et al. 1992), will provide the desired antibody specificities, or if so-called "naive libraries" in combination with in vitro afiinity maturation will offer the possibility to generate therapeutically useful human antibodies.
