Targeted Therapies for Brain Tumors: Will They Ever Deliver?

TL;DR: The neuro-oncology community is beginning to recognize that phase 0 studies should be performed to account for the impact of the blood–brain barrier on the ability of a therapeutic to reach its target(s).

Targeting BCL-2 as a Therapeutic Strategy for Primary p210BCR-ABL1-positive B-ALL Cells.

TL;DR: BCL-2 is a key survival factor for primary Ph+ B-ALL cells and its inhibition - alone or in combination with a BCR-ABL1 TKI - should be further investigated as a potential therapeutic strategy for these patients.
Journal ArticleDOI

Using genomics to guide treatment for glioblastoma.

TL;DR: As technology continues to improve, exome and RNA sequencing will play a role in the design of clinical trials, classification of patient subgroups and identification of rare mutations that can be targeted by small-molecule inhibitors and biologic agents.

The 2016 World Health Organization Classification of Tumors of the Central Nervous System: a summary.

TL;DR: The 2016 World Health Organization Classification of Tumors of the Central Nervous System is both a conceptual and practical advance over its 2007 predecessor and is hoped that it will facilitate clinical, experimental and epidemiological studies that will lead to improvements in the lives of patients with brain tumors.

CBTRUS statistical report: primary brain and central nervous system tumors diagnosed in the United States in 2007-2011.

TL;DR: The Central Brain Tumor Registry of the United States (CBTRUS), in collaboration with the Centers for Disease Control and Prevention and National Cancer Institute, is the largest population-based registry focused exclusively on primary brain and other central nervous system (CNS) tumors in the US.

Comprehensive genomic characterization defines human glioblastoma genes and core pathways

Roger E. McLendon, +233 more

TL;DR: The interim integrative analysis of DNA copy number, gene expression and DNA methylation aberrations in 206 glioblastomas reveals a link between MGMT promoter methylation and a hypermutator phenotype consequent to mismatch repair deficiency in treated gliobeasts, demonstrating that it can rapidly expand knowledge of the molecular basis of cancer.
