Abstract: Background Several elements in colostrum and human milk, including antibodies and nonspecific factors with bactericidal and antiviral activity, may play an important anti-infectious and protective role. In developing countries, enterotoxigenic Escherichia coli (ETEC) is the main etiological agent of diarrhea in low-socioeconomic level children. In the present work, we studied the functional activity of mononuclear (MN) and polymorphonuclear (PMN) phagocytes of human colostrum against ETEC, as well as the interactions between these cells and colostral or serum opsonins. Methods Colostrum samples were collected from 33 clinically healthy women between 48 and 72 hours postpartum. We verified superoxide release in colostral MN and PMN using cytochrome C reduction methods, phagocytosis, and bactericidal activity using acridine orange methods and superoxide dismutase (SOD) in the colostrum supernatants. Results Colostral MN and PMN phagocytes exposed to ETEC opsonized with colostrum supernatants caused a significant increase ( p p p Conclusions These results suggest that the ability of phagocytes to eliminate ETEC depends on the activation of cellular oxidative metabolism; moreover, activation of colostral phagocytes is likely an additional breast-feeding protection mechanism against intestinal infections in infants.

Conventional methods with chemical fixation as well as cryofixation are included, with details for subsequent steps of sample preparation to get optimal fungal specimens for visualization and examination through SEM.

Abstract: The Convention on Biological Diversity proposed the Aichi Biodiversity Targets to improve conservation policies and to balance economic development, social welfare, and the maintenance of biodiversity/ecosystem services. Brazil is a signatory of the Aichi Biodiversity Targets and is the most diverse country in terms of freshwater fish, but its national policies have supported the development of unsustainable commercial and ornamental aquaculture, which has led to serious disturbances to inland ecosystems and natural resources. We analyzed the development of Brazilian aquaculture to show how current aquaculture expansion conflicts with all 20 Aichi Targets. This case suggests that Brazil and many other megadiverse developing countries will not meet international conservation targets, stressing the need for new strategies, such as the environmental management system, to improve biodiversity conservation.

Abstract: The compounds terrein (1), butyrolactone I (2), and butyrolactone V (3) were isolated from the ethyl acetate extract (EtOAc) of the endophytic fungus Aspergillus terreus—F7 obtained from Hyptis suaveolens (L.) Poit. The extract and the compounds presented schistosomicidal activity against Schistosoma mansoni; at 100 µg/mL for EtOAc extract, 1297.3 µM for compound 1, 235.6 µM for compound 2, and 454.1 µM for compound 3, they killed 100% of the parasites after 72 h of treatment. Compounds 1, 2, and 3 exerted moderate leishmanicidal activity against Leishmania amazonensis (IC50 ranged from 23.7 to 78.6 µM). At 235.6 and 227.0 µM, compounds 2 and 3, respectively, scavenged 95.92 and 95.12% of the DPPH radical (2,2-diphenyl-1-picryl-hydrazyl), respectively. Regarding the cytotoxicity against the breast tumor cell lines MDA-MB-231 and MCF-7, compound 2 gave IC50 of 34.4 and 17.4 µM, respectively, while compound 3 afforded IC50 of 22.2 and 31.9 µM, respectively. At 117.6 µM, compound 2 inhibited the growth of and killed the pathogen Escherichia coli (ATCC 25922). Compounds 1, 2, and 3 displayed low toxicity against the normal line of human lung fibroblasts (GM07492A cells), with IC50 of 15.3 × 103, 3.4 × 103, and 5.8 × 103 µM, respectively. This is the first report on (i) the in vitro schistosomicidal and leishmanicidal activities of the EtOAc extract of A. terreus—F7 and compounds 1, 2, and 3; and (ii) the antitumor activity of compounds 2 and 3 against MDA-MB-231 and MCF-7 cells.

Abstract: Climate change poses a critical threat to the Pantanal, the largest wetland in the world. Models indicate an increase in the frequency of extreme precipitation events and extended periods of drought. These changes can amplify consequences for Pantanal's ecological functioning, which has already experienced intensive human modification of its hydrological system and environmental health. The present study analyzed the spatial and temporal dynamics of rainfall and resulting extremes in the Brazilian area of the Upper Paraguay River Basin (UPRB) along with a co-evaluation of the global Sea Surface Temperature data (SST). The predicted results indicate that wet extreme precipitation events will become more frequent in the highlands, while severe and prolonged droughts triggered by warming SSTs in the Northern Hemisphere (North Atlantic and North Pacific oceans) will affect the Pantanal. The linear relations between precipitation with SST of very specific oceanic regions and even from specific oceanic indexes obtained in the present study significantly improve the forecasting capacity, mainly from a resulting reduction to two months of the lead-time between SST warming to concomitant precipitation impacts, and by explaining 80% of Pantanal´s precipitation variation from major oceanic indexes (e.g., ENSO, PDO, NAO, ATL3). Current SST trends will result in inter- and intra-annual flooding dynamic alterations, drastically affecting the Pantanal ecosystem functioning, with consequences for wildlife diversity and distribution. Regarding the foreseeable global climate and land use change scenarios, the results from the present study provide solid evidence that can be used at different decision-making levels (from local to global) for identifying the most appropriate management practices and effectively achieving sustainability of the anthropic activity occurring in the Pantanal.
