Abstract: Obesity has been shown to impair myocardial performance. Some factors have been suggested as responsible for possible cardiac abnormalities in models of obesity, among them beta-adrenergic (βA) system, an important mechanism of regulation of myocardial contraction and relaxation. The objective of present study was to evaluate the involvement of βA system components in myocardial dysfunction induced by obesity. Thirty-day-old male Wistar rats were distributed in control (C, n = 25) and obese (Ob, n = 25) groups. The C group was fed a standard diet and Ob group was fed four unsaturated high-fat diets for 15 weeks. Cardiac function was evaluated by isolated papillary muscle preparation and βA system evaluated by using cumulative concentrations of isoproterenol and Western blot. After 15 weeks, the Ob rats developed higher adiposity index than C rats and several comorbidities; however, were not associated with changes in systolic blood pressure. Obesity caused structural changes and the myocardial responsiveness to post-rest contraction stimulus and increased extracellular calcium (Ca2+) was compromised. There were no changes in cardiac function between groups after βA stimulation. The obesity was not accompanied by changes in protein expression of G protein subunit alpha (Gsα) and βA receptors (β1AR and β2AR). In conclusion, the myocardial dysfunction caused by unsaturated high-fat diet-induced obesity, after 15 weeks, is not related to βAR system impairment at the receptor-signalling pathway.

Abstract: Summary The presence of genotype-environment interaction (GEI) influences production making the selection of cultivars in a complex process. The two most used methods to analyze GEI and evaluate genotypes are AMMI and GGE Biplot, being used for the analysis of multi environment trials data (MET). Despite their different approaches, both models complement each other in order to strengthen decision making. However, both models are based on biplots, consequently, biplot-based interpretation doesn’t scale well beyond two-dimensional plots, which happens whenever the first two components don’t capture enough variation. This paper proposes an approach to such cases based on cluster analysis combined with the concept of medoids. It also applies AMMI and GGE Biplot to the adjusted data in order to compare both models. The data is provided by the International Maize and Wheat Improvement Center (CIMMYT) and comes from the 14th Semi-Arid Wheat Yield Trial (SAWYT), an experiment concerning 50 genotypes of spring bread wheat (Triticum aestivum) germplasm adapted to low rainfall. It was performed in 36 environments across 14 countries. The analysis provided 25 genotypes clusters and 6 environments clusters. Both models were equivalent for the data’s evaluation, permitting increased reliability in the selection of superior cultivars and test environments.

Abstract: Objective To investigate antifungal susceptibility and factors associated with oral colonization by Candida species in HIV-positive patients. Methods A prospective study based on convenience sampling of subjects recruited from a pool of confirmed HIV-positive individuals seen at a specialty outpatient service in Rondonopolis, Mato Grosso, Brazil). Oral swabs were collected from 197 patients. Candida species were identified by standard microbiological techniques (phenotypic and molecular methods). Antifungal susceptibility was investigated using the broth microdilution method. Results A total of 101 (51.3%) patients were Candida spp carriers. Candida albicans was the most prevalent species (80%). Patients aged 45 to 59 years (Prevalence ratios: 1.90; 95%CI: 1.57-6.31) and 60 years or older (Prevalence ratios: 4.43; 95%CI: 1.57-34.18) were at higher risk of oral colonization by Candida species. Resistance to fluconazole and ketoconazole, or to itraconazole, corresponded to 1% and 4%, respectively. Conclusion Age (45 years or older) was the only factor associated with oral colonization by Candida . Low rates of antifungal resistance to azoles were detected in yeast isolates obtained from HIV-positive patients. Findings of this study may contribute to proper therapeutic selection for oral candidiasis in HIV-positive patients.
