Abstract: Purpose: TP53 mutations in early-stage non–small cell lung cancer (NSCLC) may be associated with worse survival but their prognostic role in advanced NSCLC is controversial. In addition, it remains unclear whether mutated patients represent a clinically homogeneous group. Experimental Design: We retrospectively examined TP53 mutations and outcome in a training cohort of 318 patients with stage IIIB–IV NSCLC: 125 epidermal growth factor receptor ( EGFR ) wild-type (wt) and 193 EGFR mutated (mut). An independent validation cohort of 64 EGFR -mut patients was subsequently analyzed. Mutations were classified as “disruptive” and “nondisruptive” according to their predicted degree of disturbance of the p53 protein structure and function. Results: In the training cohort, TP53 mutations were found in 43 of the 125 EGFR -wt patients (34.4%). Of these, 28 had nondisruptive TP53 mutations and a median overall survival (OS) of 8.5 months, compared with 15.6 months for the remaining 97 patients ( P = 0.003). In the EGFR -mut group, TP53 mutations were found in 50 of the 193 patients (25.9%). The OS for the 26 patients with TP53 nondisruptive mutations was 17.8 months versus 28.4 months for the remaining 167 patients ( P = 0.04). In the validation cohort, the 11 patients with nondisruptive TP53 mutations had a median OS of 18.1 months compared with 37.8 months for the 53 remaining patients ( P = 0.006). In multivariate analyses, nondisruptive TP53 mutations had an independent, significant association with a shorter OS. Abstract: 1. Competitive and facilitative interactions shape plant communities. Whereas a number of studies have addressed competition and direct facilitation among plants in dry ecosystems, indirect facilitation has received little attention. 2. We investigated the relative importance of direct and indirect facilitation by the nurse plant Retama sphaerocarpa on late-successional Quercus ilex seedlings mediated by herb suppression in a Mediterranean shrubland in 2006 and 2007. We also studied whether facilitation outcome depended on the size of the facilitated seedlings. 3. A field experiment was carried out to test the effect of (i) position of Q. ilex seedling with respect to shrub canopy (under shrubs or in gaps), (ii) herb competition (presence or absence), and (iii) seedling size. 2006 was an average rainfall year while 2007 had a much more humid spring and a dryer summer than 2006. 4. In both years, nurse shrubs reduced seedling mortality whereas herbs increased it. In the average rainfall year, seedling mortality under shrubs was unaffected by herbs whereas in gaps it was significantly higher in presence of herbs. This showed that the nurse shrub indirectly facilitated the seedlings by reducing the competitive capacity of herbs. Conversely, facilitation was predominately direct during the humid spring and dry summer year since herbs hindered seedling survival similarly under the nurse shrub and in gaps. The nurse shrub directly facilitated the seedlings by reducing seedling photoinhibition and water stress. 5. Improvement of environmental conditions by Retama benefited smaller seedlings but not larger seedlings since the nurse shrub reduced mortality of smaller seedlings relative to that in gaps, but this effect was not observed for larger seedlings. This indicates that individuals within a seedling population may not have the same response to facilitation. 6. Synthesis. Both indirect and direct facilitation are important mechanisms for Q. ilex regeneration in Retama shrubland and their importance seems to vary with climatic conditions. Indirect facilitation by release of herb competition under nurse shrubs is important in years of dry springs when competition between nurse shrubs and herbs is high, whereas direct facilitation mediated by microclimate amelioration increases with summer aridity.

Abstract: The aims of this study were to evaluate the clinical characteristics of HIV-negative patients affected by lymphoproliferative disorders (LPD) who developed progressive multifocal leukoencephalopathy (PML), to delineate the risk factors, and to analyze whether the new antineoplastic therapies are changing the natural history of this infectious disease. We retrospectively analyzed 46 cases with confirmed LPD-associated PML published from 1958 to 2004. Patients were stratified according to two different time periods: group A included patients diagnosed before 1989, and group B included patients diagnosed since 1990, after introduction of purine analogues. Group A patients (n = 22) had received alkylating agents and/or radiotherapy, and the majority (63.6%) had advanced Hodgkin disease. At univariate analysis, uncontrolled Hodgkin disease was the only risk factor for PML. In group B patients (n = 24), the most frequent treatments received were purine analogues (58.3%) and high-dose therapy with hematopoietic stem cell transplantation (33.3%; HDT/HSCT). B-cell chronic lymphocytic leukemia (45.8%) and aggressive non-Hodgkin lymphoma (24.9%) were the most frequent underlying LPDs. Patients treated with purine analogues were more likely to have active LPD, lower CD4 cell counts, and to be older and male than were HSCT recipients. The median interval from purine analogues or HDT/HSCT to PML was 11 months. In HDT/HSCT recipients, this interval was delayed for 10 months when peri-transplantation rituximab was used. Univariate analysis identified age >55 years, male sex, and CD4 cell counts £0.2 · 10 9 /L as risk factors for PML in patients treated with purine analogues. Mortality rates were 95.4% (group A patients), 90% (purine analogues), and 62.5% (HDT/HSCT recipients). At univariate analysis, the only factor that significantly correlated with recovery from infection was female sex. Our findings indicate (1) the possible reduction in reported cases associated with Hodgkin disease and the increasing number of published cases associated with the new antineoplastic therapies (purine analogues and HDT/ HSCT); (2) among patients treated with purine analogues, PML is more common in male patients with CD4 cell counts £0.2 · 10 9 /L; (3) the use of rituximab after HDT/ HSCT seems to delay the onset of PML; and (4) the prognosis is slightly better in transplant recipients. Abstract: Growing altricial birds may experience nutritional stress in the nest due to sibling competition, food restriction, or parasites. Nutritional stress may be detected through its effect on nestling growth, although genetic and maternal effects may interfere with its expression. A more direct way of estimating nutritional stress may be through measurements of organismic stress in growing birds. Heterophil/lymphocyte ratios (H/L) have been proposed as reliable physiological indicators of the stress response. Heat-shock protein (HSP) synthesis is induced by animals in response to various stressors, making the concentration of HSPs a suitable measure of organismic stress. In a study of growing pied flycatchers (Ficedula hypoleuca), we assessed H/L from cell counts in blood smears and levels of two HSPs, HSP60 and HSP70, from the cell fraction of peripheral blood.
