TLDR
It is proposed that this chemical selectively damages cells in the substantia nigra in patients who developed marked parkinsonism after using an illicit drug intravenously.Abstract:
Four persons developed marked parkinsonism after using an illicit drug intravenously. Analysis of the substance injected by two of these patients revealed primarily 1-methyl-4-phenyl-1,2,5,6-tetrahydropyridine (MPTP) with trace amounts of 1-methyl-4-phenyl-4-propionoxy-piperidine (MPPP). On the basis of the striking parkinsonian features observed in our patients, and additional pathological data from one previously reported case, it is proposed that this chemical selectively damages cells in the substantia nigra.read more

Maternal transmission of Alzheimer's disease: Prodromal metabolic phenotype and the search for genes
Lisa Mosconi,Valentina Berti,Russell H. Swerdlow,Alberto Pupi,Ranjan Duara,Mony J. de Leon,Mony J. de Leon +6 more
TL;DR: A review of known genetic mutations implicated in EOFAD and their effects on brain chemistry, structure and function; epidemiology and clinical research findings in LOAD, including in vivo imaging findings showing selective patterns of hypometabolism in maternally inherited AD, including chromosome X mutations, mitochondrial DNA and imprinting; and genetic mechanisms involved in other neurological disorders with known or suspected maternal inheritance as discussed by the authors.

Regulation of mitochondrial permeability transition pore by PINK1
Clement A. Gautier,Emilie Giaime,Erica Caballero,Lucía Núñez,Zhiyin Song,David C. Chan,Carlos Villalobos,Jie Shen +7 more
TL;DR: It is found that intact mitochondria in Pink1−/− cells recapitulate the respiratory defect in isolated mitochondria from PINK1−-/− mouse brains, suggesting that these PINK 1−/+ cells are a valid experimental system to study the underlying mechanisms.

Astrocytes as a primary locus for the conversion MPTP into MPP
TL;DR: Pretreating mice with fluoxetine before the administration of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine attenuated the dopaminergic neurotoxicity, consistent with the hypothesis that astrocytes are a principle site of 1%-phenylpyridinium ion production.

Animal models of Parkinson's disease: An updated overview
P. Gubellini,P. Kachidian +1 more
TL;DR: Parkinson's disease is a progressive neurodegenerative disorder whose etiology, besides a minority of genetic cases, is still largely unknown as discussed by the authors, however, the full understanding of PD is still very far from being achieved, and new potential treatments need to be tested to further improve patients' quality of life and, possibly, slow down the neuro degenerative process.

TL;DR: Biogenic amines and metabolites in the cerebrospinal fluid and microscopic evaluation of the brain at necropsy were consistent with damage to aminergic neurons in the substantia nigra.

Apraxia of lid opening
Jack E. Goldstein,David G. Cogan +1 more
TL;DR: Apraxia of lid opening is a nonparalytic motor abnormality characterized by the patient's difficulty in initiating the act of lid elevation and differs from paralytic ptosis by being present only momentarily at the start of lidOpening.
