TLDR
There is substantial evidence that ICIs are associated with a small but significant increase in risk of selected all-grade irAEs and high-grade gastrointestinal and liver toxicities, and AEs should be recognized promptly as early interventions may alleviate future complications.

Visions of the Hereafter: Releasing the Brakes of the Immune System by Checkpoint Inhibition Immunotherapy

TL;DR: Combination of different checkpoint inhibition antibodies, including novel immunoregulatory molecules may further enhance the effectiveness of this form of therapy and broaden the range of susceptible tumors.

A Case of Severe Seronegative Inflammatory Arthritis due to Nivolumab and Review of the Literature.

TL;DR: A challenging case of severe immune-related seronegative inflammatory arthritis due to nivolumab in a patient with stage IV metastatic adenocarcinoma is reported.
Journal ArticleDOI

Immune-related adverse events in cancer patients being treated with immune checkpoint inhibitors.

TL;DR: In this article, a Cox proportional hazards model was created to evaluate whether the following variables were independent predictors of an initial irAE: age ≥ 65 years, female sex, non-Caucasian race, radiation in previous 6 months, current smoking status, melanoma, and combination ICI (ipilimumab and nivolumab).

[Immune-related pneumonitis: A differential diagnosis of SARS-CoV-2 pneumonia].

TL;DR: In this paper , the authors reported the cases of three patients who presented between March and May 2020 with severe pulmonary toxicities secondary to immunotherapy, which led to with an initial hypothesis of SARS-CoV-2 pneumonia.

Commentary: Small Molecule Inhibition of PD-1 Transcription is an Effective Alternative to Antibody Blockade in Cancer Therapy.

TL;DR: The past few years has witnessed exciting progress in the application of “immune checkpoint inhibitors” (ICI) in the treatment of various human cancers through the use of antibody blockade with monoclonal antibodies that block receptor binding to their natural ligands.

Bias in meta-analysis detected by a simple, graphical test

TL;DR: Funnel plots, plots of the trials' effect estimates against sample size, are skewed and asymmetrical in the presence of publication bias and other biases Funnel plot asymmetry, measured by regression analysis, predicts discordance of results when meta-analyses are compared with single large trials.
Journal ArticleDOI

Meta-Analysis in Clinical Trials*

TL;DR: This paper examines eight published reviews each reporting results from several related trials in order to evaluate the efficacy of a certain treatment for a specified medical condition and suggests a simple noniterative procedure for characterizing the distribution of treatment effects in a series of studies.

Operating characteristics of a rank correlation test for publication bias.

TL;DR: In this paper, an adjusted rank correlation test is proposed as a technique for identifying publication bias in a meta-analysis, and its operating characteristics are evaluated via simulations, and the test statistic is a direct statistical analogue of the popular funnel-graph.
