TL;DR: The genetic architecture of schizophrenia is delineated by excluding a role for uncommon exomic variants that confer a relatively large effect (odds ratio [OR] > 4) and it is shown risk alleles within this frequency range exist, but confer smaller effects and should be identified by larger studies.

TL;DR: Studying patterns of assortative mating and anticipation, it appears increased polygenic risk is contributed by affected individuals who married into the family, resulting in an increasing genetic risk over generations, which may explain the observation of anticipation in mood disorders.
