TL;DR: In vivo combination therapy demonstrated a remarkable synergistic anti-tumor effect and prolonged survival compared to mono-therapy and the co-delivery of drug and the CSC specific inhibitor towards targeted cancer chemotherapeutics provides an insight into anticancer strategy with facile control and high efficacy.
Abstract: Cancer stem cells (CSCs) have the ability to transform into bulk cancer cells, to promote tumor growth and establish tumor metastasis. To effectively inhibit tumor growth and prevent metastasis, treatments with conventional chemotherapy drugs should be combined with CSC targeted drugs. In this study, we describe the synthesis and characterization of a new amphiphilic polymer, hyaluronic acid-cystamine-polylactic-co-glycolic acid (HA-SS-PLGA), composed of a hydrophobic PLGA head and a hydrophilic HA segment linked by a bioreducible disulfide bond. With a double emulsion method, a nano delivery system was constructed to deliver doxorubicin (DOX) and cyclopamine (CYC, a primary inhibitor of the hedgehog signaling pathway of CSCs) to both a CD44-overexpressing breast CSC subpopulation and bulk breast cancer cells and allow an on-demand release. The resulting drug-loaded NPs exhibited a redox-responsive drug release profile. Dual drug-loaded particles potently diminished the number and size of tumorspheres and HA showed a targeting effect towards breast CSCs. In vivo combination therapy further demonstrated a remarkable synergistic anti-tumor effect and prolonged survival compared to mono-therapy using the orthotopic mammary fat pad tumor growth model. The co-delivery of drug and the CSC specific inhibitor towards targeted cancer chemotherapeutics provides an insight into anticancer strategy with facile control and high efficacy.

TL;DR: Compared with several representative reducts, the proposed reduction method in incomplete decision systems can provide a mathematical quantitative measure of knowledge uncertainty and is indeed efficient, and outperforms other available approaches for feature selection from incomplete and complete data sets.
Abstract: Feature selection in large, incomplete decision systems is a challenging problem. To avoid exponential computation in exhaustive feature selection methods, many heuristic feature selection algorithms have been presented in rough set theory. However, these algorithms are still time-consuming to compute. It is therefore necessary to investigate effective and efficient heuristic algorithms. In this paper, rough entropy-based uncertainty measures are introduced to evaluate the roughness and accuracy of knowledge. Moreover, some of their properties are derived and the relationships among these measures are established. Furthermore, compared with several representative reducts, the proposed reduction method in incomplete decision systems can provide a mathematical quantitative measure of knowledge uncertainty. Then, a heuristic algorithm with low computational complexity is constructed to improve computational efficiency of feature selection in incomplete decision systems. Experimental results show that the proposed method is indeed efficient, and outperforms other available approaches for feature selection from incomplete and complete data sets.

TL;DR: A systematic investigation of the structural evolution, the electronic conduction and ionic transport properties, and the deformation pattern of lithiated SiO2 through coordinated in situ transmission electron microcopy experiments, first-principles computation, and continuum theories provides fundamental perspectives on the chemomechanical behaviors of oxides used in the surface coating of Li-ion technologies.
Abstract: Surface passivation has become a routine strategy of design to mitigate the chemomechanical degradation of high-capacity electrodes by regulating the electrochemical process of lithiation and managing the associated deformation dynamics. Oxides are the prevalent materials used for surface coating. Lithiation of SiO2 leads to drastic changes in its electro-chemo-mechanical properties from an electronic insulator and a brittle material in its pure form to a conductor and a material sustainable of large deformation in the lithiated form. We synthesized SiO2-coated SiC nanowires that allow us to focus on the lithiation behavior of the sub-10 nm SiO2 thin coating. We systematically investigate the structural evolution, the electronic conduction and ionic transport properties, and the deformation pattern of lithiated SiO2 through coordinated in situ transmission electron microcopy experiments, first-principles computation, and continuum theories. We observe the stress-mediated reaction that induces inhomogeneou...

TL;DR: The quantitative characterization provides mechanistic insight into the way the grain orientation can be engineered to mitigate the charge heterogeneity, and improves an improved understanding of the charge distribution and chemomechanical properties of polycrystalline battery materials.
Abstract: Architecting grain crystallographic orientation can modulate charge distribution and chemomechanical properties for enhancing the performance of polycrystalline battery materials. However, probing the interplay between charge distribution, grain crystallographic orientation, and performance remains a daunting challenge. Herein, we elucidate the spatially resolved charge distribution in lithium layered oxides with different grain crystallographic arrangements and establish a model to quantify their charge distributions. While the holistic "surface-to-bulk" charge distribution prevails in polycrystalline particles, the crystallographic orientation-guided redox reaction governs the charge distribution in the local charged nanodomains. Compared to the randomly oriented grains, the radially aligned grains exhibit a lower cell polarization and higher capacity retention upon battery cycling. The radially aligned grains create less tortuous lithium ion pathways, thus improving the charge homogeneity as statistically quantified from over 20 million nanodomains in polycrystalline particles. This study provides an improved understanding of the charge distribution and chemomechanical properties of polycrystalline battery materials.
