Abstract: Cylindrospermopsis raciborskii is a toxic-bloom-forming cyanobacterium that is commonly found in tropical to subtropical climatic regions worldwide, but it is also recognized as a common component of cyanobacterial communities in temperate climates. Genetic profiles of C. raciborskii were examined in 19 cultured isolates originating from geographically diverse regions of Australia and represented by two distinct morphotypes. A 609-bp region of rpoC1, a DNA-dependent RNA polymerase gene, was amplified by PCR from these isolates with cyanobacterium-specific primers. Sequence analysis revealed that all isolates belonged to the same species, including morphotypes with straight or coiled trichomes. Additional rpoC1 gene sequences obtained for a range of cyanobacteria highlighted clustering of C. raciborskii with other heterocyst-producing cyanobacteria (orders Nostocales and Stigonematales). In contrast, randomly amplified polymorphic DNA and short tandemly repeated repetitive sequence profiles revealed a greater level of genetic heterogeneity among C. raciborskii isolates than did rpoC1 gene analysis, and unique band profiles were also found among each of the cyanobacterial genera examined. A PCR test targeting a region of the rpoC1 gene unique to C. raciborskii was developed for the specific identification of C. raciborskii from both purified genomic DNA and environmental samples. The PCR was evaluated with a number of cyanobacterial isolates, but a PCR-positive result was only achieved with C. raciborskii. This method provides an accurate alternative to traditional morphological identification of C. raciborskii.

Abstract: Chronic smoking is characterized by immunosuppressive changes in the airways, leading to chronic colonization with bacteria, which in turn may contribute to the chronic obstructive pulmonary disease. The mechanisms causing this immunosuppression, however, are poorly characterized. This study evaluated whether cigarette smoke can inhibit endotoxin (LPS)-induced inflammatory cytokine production in bronchial epithelial cells and, if so, what the mechanisms are behind this effect. Pretreatment with cigarette smoke extract (CSE) concentration dependently inhibited the LPS-induced GM-CSF and IL-8 protein release, which was accompanied by decreased expression of mRNA in human bronchial epithelial cells (Beas-2B). The increase of neutrophil chemotaxis induced by conditioned medium from LPS-treated Beas-2B cells was also suppressed by CSE. In addition, the activity of LPS-induced transcription factor AP-1, but not NF-κB, was down-regulated by CSE. Notably, at the concentrations used, CSE had no effect on number or viability of Beas-2B cells. These data indicate that cigarette smoke possesses immunosuppressive properties by down-regulating the bacterial pathogen-induced neutrophil-mobilizing cytokine production via suppression of AP-1 activation in the airways. Hence, this study suggests a novel mechanism by which cigarette smoke may contribute to chronic colonization and chronic obstructive pulmonary disease in smokers.

Abstract: Precise information on the relative timing of north-south climate variations is a key to resolving ques- tions concerning the mechanisms that force and couple cli- mate changes between the hemispheres. We present a new composite record made from five well-resolved Antarctic ice core records that robustly represents the timing of re- gional Antarctic climate change during the last deglacia- tion. Using fast variations in global methane gas concen- trations as time markers, the Antarctic composite is directly compared to Greenland ice core records, allowing a de- tailed mapping of the inter-hemispheric sequence of climate changes. Consistent with prior studies the synchronized records show that warming (and cooling) trends in Antarc- tica closely match cold (and warm) periods in Greenland on millennial timescales. For the first time, we also identify a sub-millennial component to the inter-hemispheric coupling. Within the Antarctic Cold Reversal the strongest Antarctic cooling occurs during the pronounced northern warmth of the Bolling. Warming then resumes in Antarctica, potentially as early as the Intra-Allerod Cold Period, but with dating uncer- tainty that could place it as late as the onset of the Younger Dryas stadial. There is little-to-no time lag between climate transitions in Greenland and opposing changes in Antarctica. Our results lend support to fast acting inter-hemispheric cou- pling mechanisms, including recently proposed bipolar at- mospheric teleconnections and/or rapid bipolar ocean tele- connections.
