Abstract: Subjective well-being, or what is popularly often called “happiness,” has been of intense interest throughout human history. We review research showing that it is not a single factor, but that subjective well-being is composed of a number of separable although somewhat related variables. For example, positive feelings, negative feelings, and life satisfaction are clearly separable. In understanding the various types of subjective well-being, it is important to remember that appraisals move from immediate situations to a later recall of feelings, and then to global evaluations of life. At each stage, from momentary feelings to large global life eval-uations, somewhat different processes are involved in what is called “happiness.” In order to understand how to measure subjective well-being, one must understand the time course and components of the phenomenon in question, and be clear about what is most important to assess. On-line feelings are very different from global evaluations of life, although both have been studied under the rubric of subjective well-being. Although debate has focused on which type of subjective well-being should be called “true happiness,” the goal of scientists is to understand each type, their relations with each other, and their causes. The future of the field depends on understanding the differences between various types of well-being, and the different and similar causes of each.

Abstract: Plant cell walls represent the most abundant renewable resource on this planet. Despite their great abundance, only 2% of this resource is currently used by humans. Hence, research into the feasibility of using plant cell walls in the production of cost-effective biofuels is desirable. The main bottleneck for using wall materials is the recalcitrance of walls to efficient degradation into fermentable sugars. Manipulation of the wall polysaccharide biosynthetic machinery or addition of wall structure-altering agents should make it possible to tailor wall composition and architecture to enhance sugar yields upon wall digestion for biofuel fermentation. Study of the biosynthetic machinery and its regulation is still in its infancy and represents a major scientific and technical research challenge. Of course, any change in wall structure to accommodate cost-efficient biofuel production may have detrimental effects on plant growth and development due to the diverse roles of walls in the life of a plant. However, the diversity and abundance of wall structures present in the plant kingdom gives hope that this challenge can be met.

Abstract: A long-standing challenge to evolutionary theory has been whether it can explain the origin of complex organismal features. We examined this issue using digital organisms—computer programs that self-replicate, mutate, compete and evolve. Populations of digital organisms often evolved the ability to perform complex logic functions requiring the coordinated execution of many genomic instructions. Complex functions evolved by building on simpler functions that had evolved earlier, provided that these were also selectively favoured. However, no particular intermediate stage was essential for evolving complex functions. The first genotypes able to perform complex functions differed from their non-performing parents by only one or two mutations, but differed from the ancestor by many mutations that were also crucial to the new functions. In some cases, mutations that were deleterious when they appeared served as stepping-stones in the evolution of complex features. These findings show how complex functions can originate by random mutation and natural selection.

Abstract: Jasmonate (JA) is a lipid-derived hormone that regulates diverse aspects of plant immunity and development. An amino acid-conjugated form of JA, jasmonoyl–isoleucine (JA–Ile), stimulates binding of the F-box protein coronatine-insensitive 1 (COI1) to, and subsequent ubiquitin-dependent degradation of, jasmonate ZIM domain (JAZ) proteins that repress transcription of JA-responsive genes. The virulence factor coronatine (COR), which is produced by plant pathogenic strains of Pseudomonas syringae, suppresses host defense responses by activating JA signaling in a COI1-dependent manner. Although previous data indicate that COR acts as a molecular mimic of JA–Ile, the mechanism by which JA–Ile and COR are perceived by plant cells remains unknown. Here, we show that interaction of tomato COI1 with divergent members of the JAZ family is highly specific for JA–Ile and structurally related JA conjugates and that COR is ≈1,000-fold more active than JA–Ile in promoting this interaction in vitro. JA–Ile competes for binding of COR to COI1–JAZ complexes, demonstrating that COR and JA–Ile are recognized by the same receptor. Binding of COR to the COI1–JAZ complex requires COI1 and is severely impaired by a point mutation in the putative ligand-binding pocket of COI1. Finally, we show that the C-terminal region of JAZ3 containing the highly conserved Jas motif is necessary and sufficient for hormone-induced COI1–JAZ interaction. These findings demonstrate that COI1 is a critical component of the JA receptor and that COR exerts its virulence effects by functioning as a potent agonist of this receptor system.
