Abstract: Plant development is characterized by a profound ability to regenerate and form tissues with new axes of polarity. An unsolved question concerns how the position within a tissue and cues from neighboring cells are integrated to specify the polarity of individual cells. The canalization hypothesis proposes a feedback effect of the phytohormone auxin on the directionality of intercellular auxin flow as a means to polarize tissues. Here we identify a cellular and molecular mechanism for canalization. Local auxin application, wounding, or auxin accumulation during de novo organ formation lead to rearrangements in the subcellular polar localization of PIN auxin transport components. This auxin effect on PIN polarity is cell-specific, does not depend on PIN transcription, and involves the Aux/IAA-ARF (indole-3-acetic acid-auxin response factor) signaling pathway. Our data suggest that auxin acts as polarizing cue, which links individual cell polarity with tissue and organ polarity through control of PIN polar targeting. This feedback regulation provides a conceptual framework for polarization during multiple regenerative and patterning processes in plants.

Abstract: In human interactions, frequently one individual becomes 'infected' with emotions displayed by his or her partner. We tested the predictions by Hatfield et al. (1992) (Primitive emotional contagion. Review of Personal and Social Psychology 14, 151-177) that the automatic, mostly unconscious component of this process, called 'primitive emotional contagion', is repeatable and fast, that stronger facial expressions of the sender evoke stronger emotions in the viewer and that women are more susceptible to emotional contagion than men. We presented photos from the Pictures of Facial Affect (Ekman and Friesen, 1976). (Pictures of Facial Affect. Consulting Psychologists Press, Palo Alto) on a PC varying the affective content (happy and sad), the expressive strength and the duration of presentation. After each photo, subjects rated the strength of experienced happiness, sadness, anger, disgust, surprise, fear and pleasure. Feelings of happiness or sadness were significantly, specifically and repeatedly evoked in the viewer - even with presentations lasting only 500 ms. Stronger expressions evoked more emotion. The gender of the viewer had weak effects. We hypothesize that this fast and repeatable reaction is likely to have a 'prewired' neural basis. We propose that the induction of emotional processes within a subject by the perception of emotionally expressive faces is a powerful instrument in the detection of emotional states in others and as the basis for one's own reactions. Detailed knowledge of emotional reactions to faces is also valuable as a basis for psychiatric studies of disorders in affect and/or communication and in studies using functional imaging (fMRI or PET) where faces are increasingly used as stimuli.

Abstract: Leukocyte migration is a key event both in host defense against invading pathogens as well as in inflammation. Bacteria generate chemoattractants primarily by excretion (formylated peptides), complement activation (C5a), and subsequently through activation of leukocytes (e.g., leukotriene B4, platelet-activating factor, and interleukin 8). Here we describe a new protein secreted by Staphylococcus aureus that specifically impairs the response of neutrophils and monocytes to formylated peptides and C5a. This chemotaxis inhibitory protein of S. aureus (CHIPS) is a 14.1-kD protein encoded on a bacteriophage and is found in >60% of clinical isolates. CHIPS reduces the neutrophil recruitment toward C5a in a mouse peritonitis model, even though its activity is much more potent on human than on mouse cells. These findings suggest a new immune escape mechanism of S. aureus and put forward CHIPS as a potential new antiinflammatory therapeutic compound.

Abstract: Background— Stent implantation for obstructive femoropopliteal artery disease has been associated with poor long-term outcomes. This study evaluated the effectiveness of shape memory alloy recoverable technology (SMART) nitinol self-expanding stents coated with a polymer impregnated with sirolimus (rapamycin) versus uncoated SMART stents in superficial femoral artery obstructions. Methods and Results— Thirty-six patients were recruited for this double-blind, randomized, prospective trial. All patients had chronic limb ischemia and femoral artery occlusions (57%) or stenoses (average lesion length, 85±57 mm). Patients were eligible for randomization after successful guidewire passage across the lesion. Eighteen patients received sirolimus-eluting SMART stents and 18 patients received uncoated SMART stents. The primary end point of the study was the in-stent mean percent diameter stenosis, as measured by quantitative angiography at 6 months. The in-stent mean percent diameter stenosis was 22.6% in the sirolimus-eluting stent group versus 30.9% in the uncoated stent group ( P =0.294). The in-stent mean lumen diameter was significantly larger in the sirolimus-eluting stent group (4.95 mm versus 4.31 mm in the uncoated stent group; P =0.047). No serious adverse events (death or prolonged hospitalization) were reported. Conclusions— The use of sirolimus-eluting SMART stents for superficial femoral artery occlusion is feasible, with a trend toward reducing late loss compared with uncoated stents. The coated stent also proved to be safe and was not associated with any serious adverse events.
