TLDR
The most suitable conditions for protoplasting were as follows: age of the organism in slant, 3 days; mycelium age, 20 h; volume of lytic enzymes, 190 ml;Mycelial weight (dry equivalent), 1.66 g; time of contact with lytic enzyme, 2 h; temperature of protoplasts, 30°C; phosphate buffer, 25 m m , pH 6.5; KCl as osmotic stabilizer, 0.7 mAbout:

Production of β-1,3-glucanase from Trichoderma harzianum in surface and submerged culture processes and its role on protoplast generation from Trichoderma reesei mycelium
K. Théodore,Tapobrata Panda +1 more
TL;DR: It was observed that the activities of β-1,3-glucanase and carboxymethylcellulase varied during the entire fermentation cycle which had a distinct effect on protoplast generation from T. reesei mycelium.
Journal ArticleDOI
Isolation and regeneration of protoplasts from gills of Agaricus bisporus
Xiao-Ya Chen,Rüdiger Hampp +1 more
TL;DR: Investigation of specific activities of glucose-6-phosphate dehydrogenases and mannitol dehydrogenase indicated a highly active pentose phosphate pathway and a good capacity for mannitor synthesis in protoplasts, as well as in other cells of fruit-bodies of the species.
Journal ArticleDOI
Optimization of different factors for efficient protoplast release from entomopathogenic fungusMetarhizium anisopliae
Priyanka Dhar,Gurvinder Kaur +1 more
TL;DR: Mycelium harvested at 40th h in Sabouraud Dextrose broth showed the best protoplast yield over the other media and age of mycelium tested, and the most suitable osmotic stabilizer was 0.7M KCl.
Journal ArticleDOI
Penicillium citrinum protoplasts: Preparation, regeneration and lipolytic activity of regenerants
I. H. Maliszewska,E. M. Zboińska +1 more
TL;DR: The conditions for an optimum formation of protoplasts from mycelium of Pencillium citrinum, which produces extracellular lipase, were examined and some regenerants were more effective in producing lipase than the parent.

A rapid and sensitive method for the quantitation of microgram quantities of protein utilizing the principle of protein-dye binding
TL;DR: This assay is very reproducible and rapid with the dye binding process virtually complete in approximately 2 min with good color stability for 1 hr with little or no interference from cations such as sodium or potassium nor from carbohydrates such as sucrose.

A comparison of methods for estimating the viable count of a suspension of tumour cells.
TL;DR: All four in vitro methods of estimating the percentage viability of a cell suspension gave similar results with a recently harvested cell suspension but three of them grossly overestimated the proportion of viable cells in a suspension which had been stored at 4 °C for 11 days.
