TL;DR: The results suggest that the strongest selective pressures occur during early glioma development and that current therapies shape this evolution in a largely stochastic manner.

TL;DR: It is shown that evolutionary inference from integrated genomic analysis in multisector biopsies can inform targeted therapeutic interventions for patients with GBM and that targeting truncal events is more efficacious than targeting private events in reducing the tumor burden.
Journal ArticleDOI
Mutational Landscape of Secondary Glioblastoma Guides MET-Targeted Trial in Brain Tumor.
Huimin Hu,Quanhua Mu,Zhaoshi Bao,Yiyun Chen,Yanwei Liu,Jing Chen,Kuanyu Wang,Zheng Wang,Yoonhee Nam,Biaobin Jiang,Jason K. TL;DR: A MET kinase inhibitor is described, PLB-1001, that demonstrates remarkable potency in selectively inhibiting MET-altered tumor cells in preclinical models and shows blood-brain barrier permeability and is subsequently applied in a phase I clinical trial that enrolls MET- altered chemo-resistant glioma patients.
Journal ArticleDOI

TL;DR: Analysis of genomic and transcriptomic data from 462 patient-derived tumor cell (PDC) samples across 14 cancer types, along with pharmacological responses to 60 agents, indicates that PDC-derived drug sensitivities might be predictive of clinical response to targeted therapies.
Journal ArticleDOI

TL;DR: The GLASS initiative will provide essential insights into the evolution of glioma toward a lethal phenotype, with the potential to reveal targetable vulnerabilities and, ultimately, improved outcomes for a patient population in need.
