Abstract: Three molecular adducts of the antituberculosis drug isoniazid (INH) are synthesized with γ-resorcylic acid (γRA), phloroglucinol (PG), and gallic acid (GA). The new solid phases are preliminarily characterized by the thermal analysis (DSC/TGA) and powder X-ray diffraction. The formation of new solid phases is confirmed by single crystal X-ray diffraction, infrared (FT-IR) and Raman spectroscopy. All three new solid crystalline forms are stabilized by various hydrogen bonding interactions such as N+···H–O–, N···H–O, O···H–O, and π–π stacking. The FT-IR analysis puts forward that the solid form of INH1 is a salt whereas the INH2 and INH3 molecular complexes are cocrystals. We have also investigated the density of states (DOS), band structure, and atomic orbit projected density of state (PDOS) of title compounds by adopting the density functional theory (DFT) technique in the local density approximation (LDA). The electronic structure calculations show that energy states are delocalized in the k-space due the hydrogen and covalent bonds in the crystals. The frontier molecular orbital (FMO) analysis reveals that charge transfer takes place within the compounds. The Hirshfeld analysis shows that H–H and N⋯H–O hydrogen bonding interactions are dominant in all three molecular adducts of INH.

Abstract: The present study was aimed at the preparation, characterization, and evaluation of the performance of amphotericin B (AmB)-loaded aerosolized liposomes and emulsomes (core composed of solid lipid surrounded by phospholipid bilayers) for their selective presentation to lungs (alveolar macrophages), as this is the densest site of aspergillosis infection. Liposomes and emulsomes were modified by coating them with alveolar macrophage-specific ligands (O-palmitoyl mannan, OPM) and also with monoclonal antibody EBA-2. The prepared formulations were characterized in vitro for vesicle size, zeta potential and percent drug entrapment. We evaluated the therapeutic efficacy of the novel site-specific targetable lipid-based delivery systems in experimental pulmonary aspergillosis. Immunosuppressed rats with pulmonary aspergillosis were given 1 mg/kg of aerosolized liposomal and emulsomes formulations of AmB once by using an ultrasonic jet nebulizer on the third day after infection. Abstract: The present investigation reports, the bacterial cellulose (BC) a high-purity polymer produced from isolated strain ZKE7. BC production was optimized by Plackett–Burman and central composite designs yielding 18.5 g/l BC with a 4.5-fold enhancement. BC composites functionalized with antibiotics, BC-gelatin hydrogel, BC-Chitosan hydrogel and BC silver nanoparticle composites were developed and evaluated for water retention capability, moisture content and protein adsorption. The drug release behavior of the composites was consistent for controlled drug delivery. Composites were assessed for functional characteristic such as antimicrobial properties. BC composites functionalized with Neotericine exhibited antifungal activity against Candida albicans. Other composites showed pronounced antibacterial properties against Escherichia coli, Bacillus subtilis and Micrococcus luteus. Structural and thermal characterization of BC composites was carried out by FTIR, SEM with energy dispersive X-ray analysis, TGA and differential scanning calorimetry analysis. The results reveal high BC production with excellent properties that can be employed in biomedical field.
