Abstract: Dendrimers, by virtue of their therapeutic value, have recently generated enormous interest among biomedical scientists. Advancement of dendrimeric nano-architecture with well defined size, shape and controlled exterior functionality embraces promise in biomedical and pharmaceutical applications such as drug delivery, solubilization, DNA transfection and diagnosis. Highly branched, monodisperse, stable molecular level and low polydispersity with micelle-like behavior possessing nano-scale container property distinguish these structures as inimitable and optimum carrier for those applications. Dendrimers has been evaluated for delivery of different types of bioactives inside the cells. Different types of techniques are being used for exploring dendrimer safety and efficacy via cell cytotoxicity assays, cell uptake studies by fluorescent microscopy, cell line studies, flow cytometry, gamma scintigraphy and confocal microscopy; are being used over a decade. Among these, cell line studies are widely used for ex vivo characterization of dendrimers and other nanocarriers. Cell lines are homogeneous population of cells, stable after mitosis, and have an unlimited capacity for division. This review focuses on the use of different cell line studies including anticancer drugs, anti-HIV drugs, anti-inflammatory drugs, anti-tubercular-drugs, photodynamic therapy, hormonal therapy employing dendritic nanocarrier.

Abstract: Indomethacin, a non-steroidal anti-inflammatory drug was encapsulated into liposomes composed of soyabean lecithin, cholesterol and stearylamine for oral administration. Liposomes of homogenous size distribution and higher entrapment efficiency were derived from effervescent granule based proliposomes. The efficacy of the oral route was studied by measuring ulcerogenic index and anti-inflammatory activity using carrageenan induced paw oedema test in rats. The effervescent granule based liposomal products exhibited improved in vivo performance with reference to their cytoprotective and anti-inflammatory activities.

Abstract: The effect of culture medium nutrients on growth and alkaloid production by plant cell cultures of Nothapodytes nimmoniana (J. Grah.) Mabberley (Icacinaceae) was studied with a view to increasing the production of the alkaloid camptothecin, a key therapeutic drug used for its anticancer properties. Amongst the various sugars tested with Murashige and Skoog (MS) medium, such as glucose, fructose, maltose, and sucrose, maximum accumulation of camptothecin was observed with sucrose. High nitrate in the media supports the biomass, while high ammonium enhances the camptothecin content. Selective feeding of 60 mM total nitrogen with a NH4+/NO3− balance of 5/1 on day 15 of the culture cycle results in a 2.4-fold enhancement in the camptothecin content over the control culture (28.5 μg/g DW). Furthermore, the sucrose feeding strategy greatly stimulated cell biomass and camptothecin production. A modified MS medium was developed in the present study, which contained 0.5 mM phosphate, a nitrogen source feeding ratio of 50/10 mM NH4+/NO3− and 3 % sucrose with additional 2 % sucrose feeding (added on day 12 of the cell culture cycle) with 10.74 μM naphthaleneacetic acid and 0.93 μM kinetin. Finally, the selective medium has 1.7- and 2.3-fold higher intracellular and extracellular camptothecin content over the control culture (29.2 and 8.2 μg/g DW), respectively.
