Imaging biochemistry inside cells

TLDR
The development and use of sensors based on the genetically encoded variants of green-fluorescent proteins has facilitated the observation of 'live' biochemistry on a microscopic level, with the advantage of preserving the cellular context of biochemical connectivity, compartmentalization and spatial organization.

Fluorescence imaging with near-infrared light: new technological advances that enable in vivo molecular imaging.

TL;DR: FMT, a technique that can three-dimensionally image gene expression by resolving fluorescence activation in deep tissues by focusing on optical imaging technologies used for non-invasive imaging of the distribution of such probes, is discussed.
Journal ArticleDOI

The Phasor Approach to Fluorescence Lifetime Imaging Analysis

TL;DR: The phasor approach has the potential to simplify the way data are analyzed in FLIM, paving the way for the analysis of large data sets and, in general, making the FLIM technique accessible to the nonexpert in spectroscopy and data analysis.
Journal ArticleDOI

Fluorescence molecular tomography resolves protease activity in vivo

TL;DR: It is demonstrated that enzyme-activatable fluorochromes can be detected with high positional accuracy in deep tissues, that molecular specificities of different beacons towards enzymes can be resolved and that tomography of beacon activation is linearly related to enzyme concentration.
Journal ArticleDOI

Signal transduction and endocytosis: close encounters of many kinds.

TL;DR: Recent studies of protein tyrosine kinases and G-protein-coupled receptors have shed new light on the mechanisms and functional consequences of this bidirectional interplay between signalling and membrane-transport networks.

Fluorescent indicators for Ca2+based on green fluorescent proteins and calmodulin

TL;DR: New fluorescent indicators for Ca2+ that are genetically encoded without cofactors and are targetable to specific intracellular locations are constructed and dubbed ‘cameleons’.

Visualizing secretion and synaptic transmission with pH-sensitive green fluorescent proteins

TL;DR: PHluorins are developed pH-sensitive mutants of green fluorescent protein by structure-directed combinatorial mutagenesis, with the aim of exploiting the acidic pH inside secretory vesicles, to monitor vesicle exocytosis and recycling.
