Induced Pluripotent Stem Cell Neuronal Models for the Study of Autophagy Pathways in Human Neurodegenerative Disease

TL;DR: It is essential that the methodologies commonly used to study autophagy regulation under basal and stressed conditions in standard cell-line models are accurately applied when using hiPSC-derived neuronal cultures to study human neurodegenerative disease in vitro.
Journal ArticleDOI

"Miniguts" from plucked human hair meet Crohn's disease.

TL;DR: The current research project reports successful generation and characterization of induced pluripotent stem cells (iPSCs) derived from hair sheet keratinocyte cultures of a patient with Crohn's disease, and directs the patterned primitive gut tube toward intestinal organoids resembling the cellular architecture of human "miniguts".
Journal ArticleDOI

Shared mechanisms in stemness and carcinogenesis: lessons from oncogenic viruses

TL;DR: A synopsis of recent work from oncogenic viruses which contributes to this body of knowledge, establishing a nexus between infection, cancer, and stemness is presented.
Journal ArticleDOI

Mechanisms for Enhancing Cellular Reprogramming

TL;DR: This review is intended to highlight the windows of opportunities for developing mechanistically based approaches to replace the phenotypically guided methods currently employed in reprogramming, in an attempt to move the field of cell conversion towards using next generation technologies.

Induction of pluripotent stem cells from mouse embryonic and adult fibroblast cultures by defined factors.

TL;DR: Induction of pluripotent stem cells from mouse embryonic or adult fibroblasts by introducing four factors, Oct3/4, Sox2, c-Myc, and Klf4, under ES cell culture conditions is demonstrated and iPS cells, designated iPS, exhibit the morphology and growth properties of ES cells and express ES cell marker genes.
Journal ArticleDOI

Model-based Analysis of ChIP-Seq (MACS)

TL;DR: This work presents Model-based Analysis of ChIP-Seq data, MACS, which analyzes data generated by short read sequencers such as Solexa's Genome Analyzer, and uses a dynamic Poisson distribution to effectively capture local biases in the genome, allowing for more robust predictions.
Journal ArticleDOI

Core transcriptional regulatory circuitry in human embryonic stem cells.

TL;DR: Insight is provided into the transcriptional regulation of stem cells and how OCT4, SOX2, and NANOG contribute to pluripotency and self-renewal and how they collaborate to form regulatory circuitry consisting of autoregulatory and feedforward loops.
Journal ArticleDOI

Generation of induced pluripotent stem cells without Myc from mouse and human fibroblasts

TL;DR: A modified protocol for the generation of iPS cells that does not require the Myc retrovirus is described and, with this protocol, significantly fewer non-iPS background cells are obtained, and theiPS cells generated were consistently of high quality.
