The Alzheimer's disease-protective CD33 splice variant mediates adaptive loss of function via diversion to an intracellular pool.
Shoib S. Siddiqui,Stevan A. Springer,Andrea Verhagen,Venkatasubramaniam Sundaramurthy,Frederico Alisson-Silva,Weiping Jiang,Pradipta Ghosh,Ajit Varki +7 more
TL;DR: It is reported that the protection by the CD33m isoform may not be conferred by what it does but, rather, from what it cannot do, and production and localization ofCD33m in peroxisomes is a way of diminishing the amount of CD33M and enhancing β-amyloid clearance.
Journal ArticleDOI
Metabolism of Vertebrate Amino Sugars with N-Glycolyl Groups: RESISTANCE OF α2–8-LINKED N-GLYCOLYLNEURAMINIC ACID TO ENZYMATIC CLEAVAGE
Leela R. L. Davies,Oliver M. T. Pearce,Matthew B. Tessier,Siavash Assar,Victoria Smutova,Maria Pajunen,Mizuki Sumida,Chihiro Sato,Ken Kitajima,Jukka Finne,Pascal Gagneux,Alexey V. Pshezhetsky,Robert J. Woods,Robert J. Woods,Ajit Varki +14 more
TL;DR: It is confirmed that brain Neu5Gc expression has been maintained at extremely low levels over hundreds of millions of years of vertebrate evolution, and resistance of α2–8-linked Neu 5Gc to vertebrate sialidases provides a potential explanation for the rarity of Neu7Gc in the vertebrate brain.
Journal ArticleDOI
Structure of a novel sialylated fucosyl lacto-N-norhexaosylceramide isolated from chronic myelogenous leukemia cells.
TL;DR: A biosynthetic pathway of extension, sialylation, followed by fucosylation is proposed for chronic myelogenous leukemia cells based on the structures of this fucoganglioside and normal granulocyte glycolipids.
Journal ArticleDOI
Human uniqueness and the denial of death
TL;DR: Danny Brower’s contrarian view could help modify and reinvigorate ongoing debates about the origins of human uniqueness and inter-subjectivity and steer discussions of other uniquely human ‘universals’.
Journal ArticleDOI
Trans-sialidase from Trypanosoma cruzi catalyzes sialoside hydrolysis with retention of configuration
Adriane R. Todeschini,Lucia Mendonça-Previato,José O. Previato,Ajit Varki,Herman van Halbeek +4 more
TL;DR: This study shows that the stereo-selectivity of the sialid enzyme activity of T.cruzi trans -sialidase is identical to that of bacterial, viral, and mammalian sIALidases, suggesting a similar active-site architecture.
