Abstract: A system is provided for carrying out real time fluorescence-based measurements of nucleic acid amplification products. In a preferred embodiment of the invention, an excitation beam is focused into a reaction mixture through a surface, the reaction mixture containing: (i) a first fluorescent indicator capable of generating a first fluorescent signal whose intensity is proportional to the amount of an amplification product in the volume of the reaction mixture illuminated by the excitation beam and (ii) a second fluorescent indicator homogeneously distributed throughout the reaction mixture capable of generating a second fluorescent signal proportional to the volume of reaction mixture illuminated by the excitation beam. Preferably, the excitation beam is focused into the reaction mixture by a lens through a portion of a wall of a closed reaction chamber containing the reaction mixture. The same lens is used to collect the first and second fluorescent signals generated by the first and second fluorescent indicators, respectively, in response to the excitation beam. The ratio of the fluorescent intensities of the first and second fluorescent signals provides a stable quantitative indicator of the amount of amplification product synthesized in the course of the amplification reaction.

Abstract: Surani and colleagues use single-cell transcriptomics analysis in a model of mouse primordial germ cell specification to analyse the collaboration between three transcription factors, BLIMP1, PRDM14 and AP2γ, in determining germ cell fate. They find that BLIMP1 binds directly to repress somatic and cell proliferation genes, and at the same time induces AP2γ, which acts together with PRDM14. The three factors are sufficient for specification and form a tripartite interdependent transcriptional network.

Abstract: In the past 5 years, there has been a substantial increase in the use of Y-short tandem repeat loci (Y-STRs) in forensic laboratories, especially in cases where typing autosomal STRs has met with limited success. The AmpF'STR s Yfiler TM PCR amplification kit simultaneously amplifies 17 Y-STR loci including the loci in the ''European minimal haplotype'' (DYS19, DYS385a/b, DYS389I, DYS389II, DYS390, DYS391, DYS392, and DYS393), the Scientific Working Group on DNA Analysis Methods (SWGDAM) recommended Y-STR loci (DYS438 and DYS439), and the highly polymorphic loci DYS437, DYS448, DYS456, DYS458, Y GATA H4, and DYS635 (formerly known as Y GATA C4). The Yfiler TM kit was validated according to the FBI/National Standards and SWGDAM guidelines. Our results showed that full profiles are attainable with low levels of male DNA (below 125pg) and that under optimized conditions, no detectable cross-reactive products were obtained on human female DNA, bacteria, and commonly encountered animal species. Additionally, we demonstrated the ability to detect male specific profiles in admixed male and female blood samples at a ratio of 1:1000.

Abstract: . Biomass burning is the burning of living and dead vegetation. Ninety percent of all biomass-burning events are thought to be human initiated. Human induced fires are used for a variety of ''applications'' such as agricultural expansion, deforestation, bush control, weed and residue burning, and harvesting practices. Natural fires are grassland and forest fires mainly induced by lightning. It is estimated that 8700 Tg of dry matter/year are burnt each year in total. Emissions from biomass burning include a wide range of gaseous compounds and particles that contribute significantly to the tropospheric budgets on a local, regional, and even global scales. The emission of CO, CH4 and VOC affect the oxidation capacity of the troposphere by reacting with OH radicals, and emissions of nitric oxide and VOC lead to the formation of ozone and other photo oxidants. For a large number of compounds biomass burning is one of the largest single sources in the troposphere, especially in the tropics. Biomass-burning emissions play an important role in the biogeochemical cycles of carbon and nitrogen. Following the first systematic investigations on fire emissions in laboratory experiments in the 1960's, the last 20 years saw an increasing number in studies on biomass-burning emissions in various ecosystems. Recently, our knowledge of the emissions of gaseous compounds in the troposphere from fires has increased considerably. This manuscript is the first of four describing the properties biomass burning emissions. The properties of biomass-burning particles are discussed in part II and III of this review series which have been recently published, and their direct radiative effects are in part IV. This paper focuses on the review of emission ratios and emission rates of carbon monoxide, methane, volatile organics, and nitrogen containing compounds and should not be seen as a review of global emission estimates, even though we discuss the implications of our results on such studies.
