TL;DR: Review of the analytical and clinical validity as well as of the clinical utility of DNA-based testing for mutations in the four gene(s) in diagnostic, predictive and prenatal settings and for risk assessment in relatives.
Abstract: 1. Name of the disease (synonyms):
There are four subclasses of Congenital Generalized Lipodystrophy (CGL), also named Berardinelli–Seip Congenital Lipodystrophy (BSCL): Type 1 CGL (CGL1). Type 2 CGL (CGL2). Type 3 CGL (CGL3). Type 4 CGL (CGL4). 2. OMIM# of the disease:
CGL1: #608594. CGL2: #269700. CGL3: #612526. CGL4: #613327. 3. Name of the analysed genes or dna/chromosome segments:
CGL1: AGPAT2. CGL2: BSCL2. CGL3: CAV1. CGL4: PTRF. 4. OMIM# of the gene(s):
AGPAT2: *603100. BSCL2: *606158. CAV1: *601047. PTRF: *603198. Review of the analytical and clinical validity as well as of the clinical utility of DNA-based testing for mutations in the four gene(s) in ⊠ diagnostic, □ predictive and ⊠ prenatal settings and for ⊠ risk assessment in relatives.

TL;DR: An unsupervised approach to study spatial genome integrity in situ to describe previously unidentified clonal relationships is provided and the results suggest a model for how genomic instability arises in histologically benign tissue that may represent early events in cancer evolution.
Abstract: Defining the transition from benign to malignant tissue is fundamental to improve early diagnosis of cancer. Here, we provide an unsupervised approach to study spatial genome integrity in situ to gain molecular insight into clonal relationships. We employed spatially resolved transcriptomics to infer spatial copy number variations in >120 000 regions across multiple organs, in benign and malignant tissues. We demonstrate that genome-wide copy number variation reveals distinct clonal patterns within tumours and in nearby benign tissue. Our results suggest a model for how genomic instability arises in histologically benign tissue that may represent early events in cancer evolution. We highlight the power of an unsupervised approach to capture the molecular and spatial continuums in a tissue context and challenge the rationale for treatment paradigms, including focal therapy.

TL;DR: In this paper, a disk braking device overhangs a stopper with an adjusting bolt in the vertical direction, to the outside, attaching it to the lower end side of a brake lever 3A at the side receiving a leaning force in time of opening operation.
Abstract: PROBLEM TO BE SOLVED: To surely hold each individual brake lever in a symmetrically opened state in time of releasing the brake even if a gap produced by wear in brake lining is controlled so as to be automatically reduced by a lining automatic wear controlling device, at a time when this brake lining is worn out SOLUTION: A disk braking device overhangs a stopper 58 screwed with an adjusting bolt 56 in the vertical direction, to the outside, attaching it to the lower end side of a brake lever 3A at the side receiving a leaning force in time of opening operation In addition, a pedestal 59 is installed on a base plate 1 just under the stopper, and a pinion 61 is externally fitted in an upper end of the adjusting bolt 56 via a one-way clutch so as to make the bolt 56 rotatable only in a direction tightening it downward, and it is engaged with the pinion 61 with the specified backlash and a rack 57 extending in the rocking direction of the brake lever 3A is supported from the side of the base plate 1

01 Sep 2017TL;DR: The purpose of this paper is to exhaustively review contraceptive methods available in France in 2017 and to evaluate the benefits–risks balance of each of them in obese women.
