Abstract: The aim of this study was to assess the physical demands of elite English rugby union match-play. Player movements were captured by five distributed video cameras and then reconstructed on a two-dimensional plane representing the pitch. Movements based on speeds were categorized as standing, walking, jogging, and medium-intensity running (low-intensity activity), and high-intensity running, sprinting, and static exertion (scrummaging, rucking, mauling, and tackling) (high-intensity activity). Position groups were defined as forwards (tight and loose) and backs (inside and outside). Backs travelled more total distance than forwards (6127 m, s = 724 vs. 5581 m, s = 692; P < 0.05) and greater distances in walking (2351 m, s = 287 vs. 1928 m, s = 2342; P < 0.001) and high-intensity running (448 m, s = 149 vs. 298 m, s = 107; P < 0.05). Abstract: Background: In mammals, contrary to what is usually assumed, recent evidence suggests that synonymous mutations may not be selectively neutral. This position has proven contentious, not least because of the absence of a viable mechanism. Here we test whether synonymous mutations might be under selection owing to their effects on the thermodynamic stability of mRNA, mediated by changes in secondary structure. Results: We provide numerous lines of evidence that are all consistent with the above hypothesis. Most notably, by simulating evolution and reallocating the substitutions observed in the mouse lineage, we show that the location of synonymous mutations is non-random with respect to stability. Importantly, the preference for cytosine at 4-fold degenerate sites, diagnostic of selection, can be explained by its effect on mRNA stability. Likewise, by interchanging synonymous codons, we find naturally occurring mRNAs to be more stable than simulant transcripts. Housekeeping genes, whose proteins are under strong purifying selection, are also under the greatest pressure to maintain stability. Conclusion: Taken together, our results provide evidence that, in mammals, synonymous sites do not evolve neutrally, at least in part owing to selection on mRNA stability. This has implications for the application of synonymous divergence in estimating the mutation rate.

Abstract: Taguchi methods are used widely as the basis for development trials during industrial process design. Here, we describe their suitability for optimisation of the PCR. Unlike conventional strategies, these arrays revealed the effects and interactions of specific reaction components simultaneously using just a few reactions, negating the need for extensive experimental investigation. Reaction components which effected product yield were easily determined. In addition, this technique was applied to the qualitative investigation of RAPD-PCR profiles, where optimisation of the size and distribution of a number of products was determined.

Abstract: The Tiger Summit, to be hosted by Prime Minister Vladimir Putin in Russia in November 2010—the Chinese Year of the Tiger and the International Year of Biodiversity—promises to be the most significant meeting ever held to discuss the fate of a single non-human species. The Summit will culminate efforts by the Global Tiger Initiative (GTI), launched in 2008 by Robert Zoellick, World Bank President. Leaders of 13 tiger range states, supported by international donors and conservationists attending the summit, are being asked to commit to substantive measures to prevent the unthinkable: extinction of the world's last wild tiger populations. Wild tiger numbers are at an historic low. There is no evidence of breeding populations of tigers in Cambodia, China, Vietnam, and DPR Korea. Current approaches to tiger conservation are not slowing the decline in tiger numbers [1]–[3], which has continued unabated over the last two decades. While the scale of the challenge is enormous, we submit that the complexity of effective implementation is not: commitments should shift to focus on protecting tigers at spatially well-defined priority sites, supported by proven best practices of law enforcement, wildlife management, and scientific monitoring. Conflict with local people needs to be mitigated. We argue that such a shift in emphasis would reverse the decline of wild tigers and do so in a rapid and cost-efficient manner.
