Changes in the gene expression of adiponectin and glucose transporter 12 (GLUT12) in lactating and non‐lactating cows
TLDR
The results of the present study suggest that in lactating cows adiponectin plays an important role in insulin resistance in the adipose tissue; in the mammary gland, GLUT12 expression is believed to be an important factor for insulin-dependent glucose metabolism.Abstract:
Glucose delivery and uptake by the mammary gland is a rate-limiting step in milk synthesis. Insulin resistance is believed to increase throughout the body following the onset of lactation. To study glucose metabolism in peak-, late-, and non-lactating cows we analyzed the expression of an adipokine, namely, adiponectin, decreased insulin resistance, leptin, and a novel insulin-responsive glucose transporter (GLUT12) in the adipose tissue and mammary gland by using real-time polymerase chain reaction. Our results demonstrated that the mRNA level of adiponectin in the adipose tissue was greater in non-lactating cows than in peak-lactating cows. In the adipose tissue, there were no significant differences in the abundance of GLUT12 mRNA between the peak-, late-, and non-lactating cows. In contrast, in the mammary gland, the mRNA level of GLUT12 was greater in non-lactating cows than in peak- and late-lactating cows. In the adipose tissue, the mRNA level of leptin and peroxisome proliferator-activated receptor gamma 2 (PPARγ2) was greater in non-lactating cows than in peak-lactating cows. The results of the present study suggest that in lactating cows adiponectin plays an important role in insulin resistance in the adipose tissue; in the mammary gland, GLUT12 expression is believed to be an important factor for insulin-dependent glucose metabolism.read more

Adiponectin deficit during the precarious glucose economy of early lactation in dairy cows.
TL;DR: Reduced plasma adiponectin belongs to the subset of hormonal adaptations in EL dairy cows facilitating mammary glucose uptake via promotion of insulin resistance, and is associated with repression of the expression of proteins associated with the endoplasmic reticulum and involved in assembly of adiponected oligomers.

TL;DR: The findings indicate that elevation of serum adiponectin might be involved in energy metabolism just around parturition, and might exert its action through regulation of receptor expression levels in target tissues in each lactational stage in Holstein dairy cows.

TL;DR: CLA supplementation attenuated the post partum increase of circulating adiponectin thus acting towards prolongation of peripartal IR and drain of nutrients towards the mammary gland.

TL;DR: It is confirmed that obesity and type 2 diabetes are associated with low plasma adiponectin concentrations in different ethnic groups and indicate that the degree of hypoadiponectinemia is more closely related to thedegree of insulin resistance and hyperinsulinemia than to the level of adiposity and glucose intolerance.

Decreased concentration of plasma leptin in periparturient dairy cows is caused by negative energy balance.
TL;DR: The energy deficit of periparturient cows causes a sustained reduction in plasma leptin, which could benefit early lactating dairy cows by promoting a faster increase in feed intake and by diverting energy from non-vital functions such as reproduction.
