TL;DR: In this article, the authors carried out a genome-wide association study comparing 20,806 ALS cases and 59,804 controls and found that mutations predominantly in the N-terminal motor domain of KIF5A are causative for hereditary spastic paraplegia (SPG10) and Charcot-Marie-Tooth Type 2 (CMT2).

Lineage tracing: The gold standard to claim direct reprogramming in vivo.
TL;DR: The discovery that many types of fully differentiated fully differentiated somatic cells can be “reprogrammed” back to a pluripotent state through epigenetic modifications driven by specific transcription factors opened the door for cell alchemy as discussed by the authors .
