Abstract: Protein degradation is a physiological process required to maintain cellular functions. There are distinct proteolytic systems for different physiological tasks under changing environmental and pathophysiological conditions. The proteasome is responsible for the removal of oxidatively damaged proteins in the cytosol and nucleus. It has been demonstrated that proteasomal degradation increases due to mild oxidation, whereas at higher oxidant levels proteasomal degradation decreases. Moreover, the proteasome itself is affected by oxidative stress to varying degrees. The ATP-stimulated 26S proteasome is sensitive to oxidative stress, whereas the 20S form seems to be resistant. Non-degradable protein aggregates and cross-linked proteins are able to bind to the proteasome, which makes the degradation of other misfolded and damaged proteins less efficient. Consequently, inhibition of the proteasome has dramatic effects on cellular aging processes and cell viability. It seems likely that during oxidative stress cells are able to keep the nuclear protein pool free of damage, while cytosolic proteins may accumulate. This is because of the high proteasome content in the nucleus, which protects the nucleus from the formation and accumulation of non-degradable proteins. In this review we highlight the regulation of the proteasome during oxidative stress and aging.

Abstract: This paper presents a dynamic model of endogenous coalition formation in cooperative games with transferable utility (TU). The players are boundedly rational. At each time step, a player decides which of the existing coalitions to join, and demands a payoff. These decisions are determined by a best-reply rule, given the coalition structure and allocation in the previous period. Further, the players experiment with myopically suboptimal strategies whenever there are potential gains from trade. We establish an isomorphism between the set of absorbing states of the process and the set of core allocations, and show that the process converges to one of these states with probability one whenever the core is non-empty. These results do not require superadditivity of the characteristic function, and they carry over to the case of coalitional values depending on the coalition structure, and to non-transferable utility (NTU) games.

Abstract: Intense structuring of plant breeding populations challenges the design of the training set (TS) in genomic selection (GS). An important open question is how the TS should be constructed from multiple related or unrelated small biparental families to predict progeny from individual crosses. Here, we used a set of five interconnected maize (Zea mays L.) populations of doubled-haploid (DH) lines derived from four parents to systematically investigate how the composition of the TS affects the prediction accuracy for lines from individual crosses. A total of 635 DH lines genotyped with 16,741 polymorphic SNPs were evaluated for five traits including Gibberella ear rot severity and three kernel yield component traits. The populations showed a genomic similarity pattern, which reflects the crossing scheme with a clear separation of full sibs, half sibs, and unrelated groups. Prediction accuracies within full-sib families of DH lines followed closely theoretical expectations, accounting for the influence of sample size and heritability of the trait. Prediction accuracies declined by 42% if full-sib DH lines were replaced by half-sib DH lines, but statistically significantly better results could be achieved if half-sib DH lines were available from both instead of only one parent of the validation population. Once both parents of the validation population were represented in the TS, including more crosses with a constant TS size did not increase accuracies. Unrelated crosses showing opposite linkage phases with the validation population resulted in negative or reduced prediction accuracies, if used alone or in combination with related families, respectively. We suggest identifying and excluding such crosses from the TS. Moreover, the observed variability among populations and traits suggests that these uncertainties must be taken into account in models optimizing the allocation of resources in GS.

Abstract: The causative agents of many infectious diseases are excreted by the faecal route and also with other excretions or secretions of the body. Some pathogens are also excreted from clinically healthy animals, from those with latent infections and in cases of transmissible multifactorial diseases. In all types of livestock housing, the pathogens finally reach the floor with the installations for collecting manure as a solid or liquid. Under these conditions livestock owners do not realise that manure may contain pathogens, and therefore do not take precautions against possible spread of diseases by utilisation of manure. The pathogens do not survive very long in stored farmyard manure because of the temperatures and biological and biochemical activities prevailing in the middens. But the conditions in slurry are different because the temperature does not rise and biochemical activity is low. Therefore the pathogens survive for rather long periods in slurry. To avoid disease transfer by utilisation of manure and slurry as fertilisers, certain precautions are necessary and these are described in detail. The agricultural utilisation of municipal sewage sludge is common in many countries. However, these sludges contain pathogens which are excreted by the human population served by the sewers and sewage treatment plants. In the sewage purification processes most of the pathogens are reduced in number but not completely eliminated. They are enriched by sedimentation processes in the sewage sludge. To protect the livestock of farms utilising sewage sludge as fertiliser or for amending soils it is necessary to sanitise hygienically dubious sludges prior to their use. The epidemiological aspects of agricultural sludge utilisation are discussed and details of the available sanitation technologies are given.
