TLDR
A stem cell associated with the bone marrow has epithelial cell lineage capability and a proportion of the regenerated hepatic cells were shown to be donor-derived.Abstract:Ā
Bone marrow stem cells develop into hematopoietic and mesenchymal lineages but have not been known to participate in production of hepatocytes, biliary cells, or oval cells during liver regeneration. Cross-sex or cross-strain bone marrow and whole liver transplantation were used to trace the origin of the repopulating liver cells. Transplanted rats were treated with 2-acetylaminofluorene, to block hepatocyte proliferation, and then hepatic injury, to induce oval cell proliferation. Markers for Y chromosome, dipeptidyl peptidase IV enzyme, and L21-6 antigen were used to identify liver cells of bone marrow origin. From these cells, a proportion of the regenerated hepatic cells were shown to be donor-derived. Thus, a stem cell associated with the bone marrow has epithelial cell lineage capability.read more

TL;DR: It is reported here that liver-specific deletion of the neurofibromatosis type 2 (Nf2) tumor suppressor gene in the developing or adult mouse specifically yields a dramatic, progressive expansion of progenitor cells throughout the liver without affecting differentiated hepatocytes.

Hematopoietic myelomonocytic cells are the major source of hepatocyte fusion partners.
TL;DR: It is demonstrated that the hematopoietic progeny of a single HSC is sufficient to produce functional hepatic repopulation and the possibility that differentiated myeloid cells may be useful for future therapeutic applications of in vivo cellular fusion is raised.

TL;DR: The present data confirm previous reports suggesting that MSCs have immunomodulatory properties in vitro, but their tolerogenic properties in vivo must be questioned, as MSC injections were not only ineffective at prolonging allograft survival, but tended to promote rejection.

Stem Cells: Hype and Reality
TL;DR: This update discusses what is known regarding embryonic and adult tissue-derived pluripotent stem cells, including the mechanisms underlying self-renewal without senescence, differentiation in multiple cell types both in vitro and in vivo, and future potential clinical uses of such stem cells.

Expression of stem cell factor and its receptor, c-kit, during liver regeneration from putative stem cells in adult rat.
TL;DR: The SCF/c-kit system may, possibly in combination with other growth factor/receptor systems, be involved in the early activation of the hepatic stem cells as well as in the expansion and differentiation of oval cells.

TL;DR: Both the GVHD propensity and tolerogenicity in these experiments were closely associated with recipient tissue chimerism 30 and 100 days after the experiments began, and these observations provide guidelines that should be considered in devising leukocyte augmentation protocols for human whole organ recipients.
