TLDR
A paradigm shift in the treatment of selected B-cell malignancies is anticipated, moving from targeting primarily the malignant cells toward combining cytotoxic drugs with agents that interfere with the microenvironment's proactive role.About:

Overview of recent developments in chronic lymphocytic leukemia.
Preetesh Jain,Kanti R. Rai +1 more
TL;DR: Prognostication and therapy in CLL have come a long way in the past three decades since the development of Rai and Binet clinical staging systems, with chimeric antigen receptor therapy based on autologous T-lymphocytes the most recent advance.
Journal ArticleDOI
The role of the CCN family of proteins in blood cancers
TL;DR: The CCN family of proteins have been recognised to play a key role in all aspects of haematopoiesis and the associated cell communication mechanisms are thus crucial to the development of new treatment strategies.

Modelling chronic lymphocytic leukaemia (CLL) cell : microenvironment interactions in two- and three-dimensional culture systems
TL;DR: The use of a novel method, ‘acoustic trapping’ to model the CLL microenvironment is investigated and results demonstrate that the HFFF2 Cell line is a suitable model for investigating CLL/microenvironment interactions and in this Model H FFF2 cells, accessory cells and soluble factors are working together to provide Pro-survival signals to the C LL cells.
Book ChapterDOI
Dysregulation of Apoptosis and Proliferation in CLL Cells
TL;DR: The aim of this chapter is to discuss essential abnormalities of CLL cells apoptosis and proliferation which contribute to the development of the disease and may determine its clinical course.
Journal ArticleDOI
Expression of far upstream element binding protein 1 in B‑cell non‑Hodgkin lymphoma is correlated with tumor growth and cell‑adhesion mediated drug resistance.
Yuejiao Huang,Xiaohong Xu,Lili Ji,Yuchan Wang,Shitao Wang,Jie Tang,Xianting Huang,Xiaojing Yang,Yunhua He,Song He,Chun Cheng +10 more
TL;DR: Findings support a role for FBP1 in non‑Hodgkin lymphoma cell proliferation, adhesion and drug resistance, and may lead to the generation of a novel therapeutic approach targeting this molecule.

Stromal Fibroblasts Present in Invasive Human Breast Carcinomas Promote Tumor Growth and Angiogenesis through Elevated SDF-1/CXCL12 Secretion
Akira Orimo,Piyush Gupta,Dennis C. Sgroi,Fernando Arenzana-Seisdedos,Thierry Delaunay,Rizwan Naeem,Vincent J. Carey,Andrea L. Richardson,Robert A. Weinberg +8 more
TL;DR: Using a coimplantation tumor xenograft model, it is demonstrated that carcinoma-associated fibroblasts extracted from human breast carcinomas promote the growth of admixed breast carcinoma cells significantly more than do normal mammaries derived from the same patients.

Macrophages: Obligate Partners for Tumor Cell Migration, Invasion, and Metastasis
TL;DR: Macrophages within the tumor microenvironment facilitate angiogenesis and extracellular-matrix breakdown and remodeling and promote tumor cell motility and are an important drug target for cancer therapy.

Conditions controlling the proliferation of haemopoietic stem cells in vitro.
TL;DR: A liquid culture system is described whereby proliferation of haemopoietic stem cells, production of granulocyte precursor cells (CFU‐C), and extensive granulopoiesis can be maintained in vitro for several months.

Maintenance of the Hematopoietic Stem Cell Pool by CXCL12-CXCR4 Chemokine Signaling in Bone Marrow Stromal Cell Niches
TL;DR: It is shown that the induced deletion of CXCR4, a receptor for CXC chemokine ligand (CXCL) 12 in adult mice, resulted in severe reduction of HSC numbers and increased sensitivity to myelotoxic injury, although it did not impair expansion of the more mature progenitors.
