TL;DR: Methods for the large-scale analysis of genetic variation in Plasmodium falciparum by deep sequencing of parasite DNA obtained from the blood of patients with malaria, either directly or after short-term culture are described.
Journal ArticleDOI

TL;DR: In this article, the authors analyzed the K13-propeller sequence polymorphism in 14,037 samples collected in 59 countries in which malaria is endemic and identified 108 nonsynonymous K13 mutations, which showed marked geographic disparity in their frequency and distribution.
Journal ArticleDOI
Two Nonrecombining Sympatric Forms of the Human Malaria Parasite Plasmodium ovale Occur Globally
Colin J. Sutherland,Naowarat Tanomsing,Debbie Nolder,Mary C. Oguike,Charlie Jennison,Sasithon Pukrittayakamee,Christiane Dolecek,Tran Tinh Hien,Virgílio E. do Rosário,Ana Paula Arez,João Pinto,Pascal Michon,Ananias A. Day,Nicholas P. J. Day,Georges Snounou,Georges Snounou,Peter J. Lockhart,Peter L. Chiodini,Mallika Imwong,Spencer D. Polley +31 more
TL;DR: It is proposed that P. ovale comprises 2 nonrecombining species that are sympatric in Africa and Asia and speculate on possible scenarios that could have led to this speciation.
Journal ArticleDOI
Plasmodium falciparum associated with severe childhood malaria preferentially expresses PfEMP1 encoded by group A var genes.
Anja T. R. Jensen,Pamela Magistrado,Sarah Sharp,Louise Joergensen,Thomas Lavstsen,Antonella Chiucchiuini,Ali Salanti,Lasse S Vestergaard,John Lusingu,Rob Hermsen,Robert W. Sauerwein,Jesper Christensen,Morten Nielsen,Lars Hviid,Colin J. Sutherland,Trine Staalsoe,Thor G. Theander +16 more
TL;DR: The proteins encoded by Group A var genes, such as PFD1235w/MAL7P1.1, appear to be involved in the pathogenesis of severe disease and are thus attractive candidates for a vaccine against life-threatening P. falciparum malaria.
Journal ArticleDOI
Highly Sensitive Detection of Malaria Parasitemia in a Malaria-Endemic Setting: Performance of a New Loop-Mediated Isothermal Amplification Kit in a Remote Clinic in Uganda
Heidi Hopkins,Iveth J. González,Spencer D. Polley,Patrick Angutoko,John Ategeka,Caroline Asiimwe,Bosco Agaba,Daniel J Kyabayinze,Colin J. Sutherland,Mark D. Perkins,David Bell +10 more
TL;DR: Malaria LAMP dramatically lowers the detection threshold achievable in malaria-endemic settings, providing a new tool for diagnosis, surveillance, and screening in elimination strategies.
