TL;DR: The population comparisons confirmed that most of the HLA-G variability has originated before human dispersion from Africa and that the allele and haplotype frequencies have probably been shaped by strong selective pressures.
Abstract: Human Leucocyte Antigen G (HLA-G) belongs to the family of nonclassical HLA class I genes, located within the major histocompatibility complex (MHC). HLA-G has been the target of most recent research regarding the function of class I nonclassical genes. The main features that distinguish HLA-G from classical class I genes are: a) limited protein variability; b) alternative splicing generating several membrane bound and soluble isoforms; c) short cytoplasmic tail; d) modulation of immune response (immune tolerance); e) restricted expression to certain tissues. In the present work, we describe the HLA-G gene structure and address the HLA-G variability and haplotype diversity among several populations around the world, considering each of its major segments (promoter, coding and 3’untranslated regions). For this purpose, we developed a pipeline to reevaluate the 1000Genomes data and recover miscalled or missing genotypes and haplotypes. It became clear that the overall structure of the HLA-G molecule has been maintained during the evolutionary process and that most of the variation sites found in the HLA-G coding region are either coding synonymous or intronic mutations. In addition, only a few frequent and divergent extended haplotypes are found when the promoter, coding and 3’ untranslated regions are evaluated together. The divergence is particularly evident for the regulatory regions. The population comparisons confirmed that most of the HLA-G variability has originated before human dispersion from Africa and that the allele and haplotype frequencies have probably been shaped by strong selective pressures.

TL;DR: A new biotin derivative functionalized by an electropolymerizable pyrrole group has been synthesized and the electrooxidation of this biotin pyr role has allowed the formation of biotinylated conducting polypyrrole films in organic electrolyte.
Abstract: A new biotin derivative functionalized by an electropolymerizable pyrrole group has been synthesized. The electrooxidation of this biotin pyrrole has allowed the formation of biotinylated conducting polypyrrole films in organic electrolyte. Gravimetric measurements based on a quartz crystal microbalance, modified by the biotinylated polymer, revealed an avidin−biotin-specific binding at the interface of polymer−solution. The estimated mass increase corresponded to the anchoring of 1.5 avidin monolayers on the polypyrrole surface. In addition, the subsequent grafting of biotinylated glucose oxidase was corroborated by electrochemical permeation studies. Enzyme multilayers composed of glucose oxidase or polyphenol oxidase were elaborated on the electrode surface modified by the biotinylated polypyrrole film. The amperometric response of the resulting biosensors to glucose or catechol has been studied at +0.6 or −0.2 V vs SCE, respectively.

TL;DR: These findings support the contention that serotonin and GABAergic neurons may act in concert in the modulation of defense reaction in the midbrain tectum, and indicate a direct neural pathway connecting the dorsal midbrain and monoaminergic nuclei of the descending pain inhibitory system.

TL;DR: Resistance mechanisms in fluconazole-resistant isolates of C. albicans isolated from AIDS patients from nine Brazilian hospitals include the presence of point mutations in the ERG11 gene and overexpression of ERG 11, and several genes encoding efflux pumps, as measured by quantitative real-time reverse transcriptase polymerase chain reaction.
