Abstract: The effect of ethanol on exponential phase cultures of S. cerevisiae has been examined using l-alanine uptake and proton efflux as indices of ethanol tolerance. Preincubation with 2 M ethanol inhibited l-alanine uptake, proton efflux and fermentation rates. However, the effect of ethanol varied in yeast cells enriched with different fatty acyl residues. It was observed that cells enriched with polyunsaturated fatty acids acquired greater tolerance to ethanol as compared to monounsaturated fatty acids. By varying the degree of unsaturation of supplemented fatty acid, a sequential insertion of double bonds in yeast membrane lipid was achieved. Results demonstrated that S. cerevisiae became more resistant to ethanol with an increase in the degree of unsaturation and that membrane fluidity could be an important determinant of ethanol tolerance.

Abstract: Conflicts between local people and protected area managers are a common problem in developing countries, but in many cases there has been little attempt to comprehensively characterize the underlying problems. Resource uses, management practices, economy and people's perceptions of problems and likely solutions were analysed in two villages near and two villages away from the core zone of Nanda Devi Biosphere Reserve in the Indian Himalaya. Agriculture, although practised on less than 1% of the area, was the primary occupation of local people. Six annual crops of a total of 22 and all four horticultural crops on private farms were damaged by wildlife, but Reserve management provided compensation only for livestock killing by wildlife and compensation amounted to only 4–10% of the total assessed monetary value of killed livestock. A variety of wild plant products were used locally but 27 were marketed by more than 50% of surveyed families; income from wild products was substantially lower than that from crops and livestock. A sociocultural change from a subsistence to a market economy, together with changes in traditional land/resource rights and institutions, has led to a number of changes in land-use and management practices. The livestock population has declined, agricultural area has remained the same and people have started cultivating medicinal species in the last 20 years. These changes seem complementary to the goal of conservation. However, changes such as abandonment of some traditional food crops and stress on cash crops lacking fodder value, requiring substantial manure inputs derived from forest litter and livestock excreta, and causing severe soil erosion, seem to counter the goal of environmental conservation. Some government-managed Reserve Forest sites were similar to the Community Forests in terms of species richness, basal area and soil physico-chemical properties. Two Reserve Forest sites showed basal areas of 160.5–191.5 m2/ha, exceeding the highest values reported so far from the region. The formal institutional framework of resource management seems to be not as effective as the traditional informal system. The Reserve Management Plan lays more emphasis on legal protection than on the sustainable livelihood of local communities and has led to conflicts between local people and reserve managers. Plantation of fodder and medicinal species in degraded forest lands, suppression of economic exploitation of local people in the market, enhancement of local knowledge of the economic potential of biodiversity, incentives for cultivation of crops with comparative advantages and lesser risks of damage by wildlife, and rejuvenation of the traditional involvement of the whole village community in decison-making, could be the options for resolving conflicts between people and protected areas in this case.

Abstract: This study was undertaken to investigate the influence of plant probiotic fungus Piriformospora indica on the medicinal plant C. forskohlii. Interaction of the C. forskohlii with the root endophyte P. indica under field conditions, results in an overall increase in aerial biomass, chlorophyll contents and phosphorus acquisition. The fungus also promoted inflorescence development, consequently the amount of p-cymene in the inflorescence increased. Growth of the root thickness was reduced in P. indica treated plants as they became fibrous, but developed more lateral roots. Because of the smaller root biomass, the content of forskolin was decreased. The symbiotic interaction of C. forskohlii with P. indica under field conditions promoted biomass production of the aerial parts of the plant including flower development. The plant aerial parts are important source of metabolites for medicinal application. Therefore we suggest that the use of the root endophyte fungus P. indica in sustainable agriculture will enhance the medicinally important chemical production.

Abstract: Groundwater evaluation indices, multivariate statistical techniques, and geostatistical models are applied to assess the source apportionment and spatial variability of groundwater pollutants at the Lakshimpur district of Bangladesh. A total of 70 groundwater samples have been collected from wells (shallow to deep wells, i.e., 10–375 m) from the study area. Groundwater quality index reveals that 50 % of the water samples belong to good-quality water. The degrees of contamination, heavy metal pollution index, and heavy metal evaluation index present diversified results in samples even though they show significant correlations among them. The results of principal component analysis (PCA) show that groundwater quality in the study area mainly has geogenic (weathering and geochemical alteration of source rock) sources followed by anthropogenic source (agrogenic, domestic sewage, etc.). Cluster analysis and correlation matrix also supported the results of PCA. The Gaussian semivariogram models have been tested as the best fit models for most of the water quality indices and PCA components. The results of semivariogram models have shown that most of the variables have weak spatial dependence, indicating agricultural and residential/domestic influences. The spatial distribution maps of water quality parameters have provided a useful and robust visual tool for decision makers toward defining adaptive measures. This study is an implication to show the multiple approaches for quality assessment and spatial variability of groundwater as an effort toward a more effective groundwater quality management.

Abstract: Hydrogen peroxide (H2O2) is known to induce cell cycle arrest and apoptosis in various somatic cell types cultured in vitro. We hypothesize that this reactive oxygen species (ROS) could modulate cell cycle and induce morphological features characteristics of apoptosis in oocytes cultured in vitro. To test this hypothesis, immature and mature oocytes were cultured in medium containing various doses of H2O2 with or without caspase-3 inhibitor for various times. The treatment of H2O2 induced germinal vesicle break down (GVBD) in all immature oocytes followed by initiation of shrinkage. Some of immature oocytes (but not mature oocytes) also showed membrane blebbing. On the other hand, H2O2 treatment inhibited first polar body emission in mature oocytes just prior to initiation of shrinkage. The cytoplasmic granulation and fragmentation into apoptotic bodies were observed in mature oocytes during later stages of H2O2 treatment. The shrinkage was induced by H2O2 in a dose- and time-dependent manner in both immature and mature oocytes. Although, H2O2-induced degeneration was observed in both immature and mature oocytes after 2.0 hrs of treatment, immature oocytes were more susceptible to undergo quick shrinkage, membrane blebbing and degeneration. Co-addition of caspase-3 inhibitor prevented shrinkage and degeneration of both immature and mature oocytes except membrane blebbing that was observed at higher doses of H2O2 after 1.0 hr of culture. Treatment of H2O2 induced bax protein expression (3 times), DNA fragmentation and caspase-3 activity (2.5 times) in oocytes undergoing morphological apoptotic changes. These findings clearly suggest that H2O2 induced GVBD in immature oocytes, inhibited first polar body extrusion in mature oocytes prior to initiation of morphological changes characteristic of apoptosis such as shrinkage, membrane blebbing and cytoplasmic fragmentation prior to degeneration.
