Abstract: Iodoacetamide (IAA) and iodoacetate (IA) have frequently been used to inhibit glycolysis, since these compounds are known for their ability to irreversibly inhibit the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH). However, the consequences of a treatment with such thiol reagents on the glutathione (GSH) metabolism of brain cells have not been explored. Exposure of astroglia-rich primary cultures to IAA or IA in concentrations of up to 1 mM deprived the cells of GSH, inhibited cellular GAPDH activity, lowered cellular lactate production and caused a delayed cell death that was detectable after 90 min of incubation. However, the two thiol reagents differed substantially in their potential to deprive cellular GSH and to inhibit astrocytic glycolysis. IAA depleted the cellular GSH content more efficiently than IA as demonstrated by half-maximal effects for IAA and IA that were observed at concentrations of about 10 and 100 muM, respectively. In contrast, IA was highly efficient in inactivating GAPDH and lactate production with half-maximal effects observed already at a concentration below 100 muM, whereas IAA had to be applied in 10 times higher concentration to inhibit lactate production by 50%. These substantial differences of IAA and IA to affect GSH content and glycolysis of cultured astrocytes suggest that in order to inhibit astrocytic glycolysis without substantially compromising the cellular GSH metabolism, IA - and not IAA - should be used in low concentrations and/or for short incubation periods.

Abstract: The tertiary sulphonium compound, β-dimethylsulphoniopropionate (DMSP) and the quaternary ammonium compounds glycine betaine and homarine are important osmotica in Platymonas subcordiformis cells. Following hypersaline stresses the compounds were accumulated after a lag period of 3 h and equilibrium concentrations were reached 6 h later. In contrast to these organic solutes, mannitol was synthesised immediately and equilibrium concentrations were reached within 90 min. Hyposaline stresses induced losses of the organic solutes from the cells. The ions K+, Na+, Cl- and the above organic solutes can account for the osmotic balance of the cells.

Abstract: Inline digital holograms are classically reconstructed using linear operators to model diffraction. It has long been recognized that such reconstruction operators do not invert the hologram formation operator. Classical linear reconstructions yield images with artifacts such as distortions near the field-of-view boundaries or twin images. When objects located at different depths are reconstructed from a hologram, in-focus and out-of-focus images of all objects superimpose upon each other. Additional processing, such as maximum-of-focus detection, is thus unavoidable for any successful use of the reconstructed volume. In this Letter, we consider inverting the hologram formation model in a Bayesian framework. We suggest the use of a sparsity-promoting prior, verified in many inline holography applications, and present a simple iterative algorithm for 3D object reconstruction under sparsity and positivity constraints. Preliminary results with both simulated and experimental holograms are highly promising.

Abstract: Much of the current work on model transformations seems essentially operational and executable in nature. Executable descriptions are necessary from the point of view of implementation. But from a conceptual point of view, transformations can also be viewed as descriptive models by stating only the properties a transformation has to fulfill and by omitting execution details. This contribution discusses the view that model transformations can be abstracted as being transformation models. As a simple example for a transformation model, the well-known transformation from the Entity-Relationship model to the Relational model is shown. A transformation model in this contribution is nothing more than an ordinary, simple model, i.e., a UML/MOF class diagram together with OCL constraints. A transformation model may transport syntax and semantics of the described domain. The contribution thus covers two views on transformations: An operational model transformation view and a descriptive transformation model view.
