Abstract: The aim of this study was to examine the capability of structure-from-motion photogrammetry in defining the geometry of cliffs and undercuts in rocks of complex geomorphology. A case site was chosen along pocket beaches near the village of Stara Baska on the Adriatic Sea island of Krk, Gulf of Kvarner, Croatia, where cliff erosion of 5 m in breccias was identified by comparison of aerial photographs from 1960 and 2004. The 3D point cloud was derived from approx. 800 photos taken on 9 January 2014 by a single camera from various elevations and angles, and processed using the online software ReCap (Autodesk). Data acquisition was found to be quick and the method easy to implement. The difference between the georeferenced 3D cloud points and an RTK-GPS survey was 7 cm, i.e. within the limits of RTK-GPS precision. Quantifying the spatial variation in undercut geometries revealed that the deepest and largest (17 m3) undercut was in the south-eastern sector of the beach. Reconstructing the detailed geomorphology of this 3.8-m-deep undercut convincingly demonstrates the high efficiency of the method. Such assessments of spatiotemporal changes in undercut and overhang volumes can prove useful for evaluations of cliff erosion risk. Coupled with the low cost and relatively simple application, this is evidently an attractive technique for meaningful geotechnical and coastal engineering monitoring in the future on the island of Krk and, for that matter, also on other Adriatic islands and in similar settings worldwide.

Abstract: Botulinum neurotoxins (BoNTs) are the most lethal toxin known to human. Biodefense requires early and rapid detection of BoNTs. Traditionally, BoNTs can be detected by looking for signs of botulism in mice that receive an injection of human material, serum or stool. While the living animal assay remains the most sensitive approach, it is costly, slow and associated with legal and ethical constrains. Various biochemical, optical and mechanical methods have been developed for BoNTs detection with improved speed, but with lesser sensitivity. Here, we report a novel nanopore-based BoNT type B (BoNT-B) sensor that monitors the toxin’s enzymatic activity on its substrate, a recombinant synaptic protein synaptobrevin 2 derivative. By analyzing the modulation of the pore current caused by the specific BoNT-B-digested peptide as a marker, the presence of BoNT-B at a subnanomolar concentration was identified within minutes. Abstract: We present a study of the very high energy (VHE; E > 100 GeV) gamma-ray emission of the blazar PKS 1424+240 observed with the MAGIC telescopes. The primary aim of this paper is the multiwavelength spectral characterization and modeling of this blazar, which is made particularly interesting by the recent discovery of a lower limit of its redshift of z > 0.6 and makes it a promising candidate to be the most distant VHE source. The source has been observed with the MAGIC telescopes in VHE gamma rays for a total observation time of ~33.6 h from 2009 to 2011. The source was marginally detected in VHE gamma rays during 2009 and 2010, and later, the detection was confirmed during an optical outburst in 2011. The combined significance of the stacked sample is ~7.2 sigma. The differential spectra measured during the different campaigns can be described by steep power laws with the indices ranging from 3.5 +/- 1.2 to 5.0 +/- 1.7. The MAGIC spectra corrected for the absorption due to the extragalactic background light connect smoothly, within systematic errors, with the mean spectrum in 2009-2011 observed at lower energies by the Fermi-LAT. The absorption-corrected MAGIC spectrum is flat with no apparent turn down up to 400 GeV. The multiwavelength light curve shows increasing flux in radio and optical bands that could point to a common origin from the same region of the jet. The large separation between the two peaks of the constructed non-simultaneous spectral energy distribution also requires an extremely high Doppler factor if an one zone synchrotron self-Compton model is applied. We find that a two-component synchrotron self-Compton model describes the spectral energy distribution of the source well, if the source is located at z~0.6.

Abstract: High throughput sequencing (HTS) generates large amounts of high quality sequence data for microbial genomics. The value of HTS for microbial forensics is the speed at which evidence can be collected and the power to characterize microbial-related evidence to solve biocrimes and bioterrorist events. As HTS technologies continue to improve, they provide increasingly powerful sets of tools to support the entire field of microbial forensics. Accurate, credible results allow analysis and interpretation, significantly influencing the course and/or focus of an investigation, and can impact the response of the government to an attack having individual, political, economic or military consequences. Interpretation of the results of microbial forensic analyses relies on understanding the performance and limitations of HTS methods, including analytical processes, assays and data interpretation. The utility of HTS must be defined carefully within established operating conditions and tolerances. Validation is essential in the development and implementation of microbial forensics methods used for formulating investigative leads attribution. HTS strategies vary, requiring guiding principles for HTS system validation. Three initial aspects of HTS, irrespective of chemistry, instrumentation or software are: 1) sample preparation, 2) sequencing, and 3) data analysis. Criteria that should be considered for HTS validation for microbial forensics are presented here. Validation should be defined in terms of specific application and the criteria described here comprise a foundation for investigators to establish, validate and implement HTS as a tool in microbial forensics, enhancing public safety and national security.
