Isolation and Characterization of Mononuclear Phagocytes in the Mouse Lung and Lymph Nodes
TLDR
How to identify pulmonary MPs using flow cytometry and how to isolate them via cell sorting are described and will be important to understanding their role in maintaining homeostasis and during the development of disease.Abstract:
There is a diverse population of mononuclear phagocytes (MPs) in the lungs, comprised of macrophages, dendritic cells (DCs), and monocytes. The existence of these various cell types suggests that there is a clear division of labor and delicate balance between the MPs under steady-state and inflammatory conditions. Here we describe how to identify pulmonary MPs using flow cytometry and how to isolate them via cell sorting. In steady-state conditions, murine lungs contain a uniform population of alveolar macrophages (AMs), three distinct interstitial macrophage (IM) populations, three DC subtypes, and a small number of tissue-trafficking monocytes. During an inflammatory response, the monocyte population is more abundant and complex since it acquires either macrophage-like or DC-like features. All in all, studying how these cell types interact with each other, structural cells, and other leukocytes within the environment will be important to understanding their role in maintaining homeostasis and during the development of disease.read more

TL;DR: It is shown across multiple murine tumor and metastasis models that CCR2 antagonism in combination with anti- PD-1 therapy leads to sensitization and enhanced tumor response over anti-PD-1 monotherapy and that enhanced treatment response correlates with enhanced CD8 + T cell recruitment and activation and a concomitant decrease in CD4 + regulatory T cell.

TL;DR: This cross-species expression compendium serves as a resource for future translational studies to investigate beforehand whether pursuing specific MP subtypes or genes will prove fruitful, and only 13%–23% of the top 1,000 marker genes overlap in corresponding human-mouse MP counterparts.

TL;DR: The data indicate a role for IL-1R as a regulator of host homeostasis and point to cachexia as a cost of long-term reliance on IL- 1–mediated tolerance mechanisms.

TL;DR: A fate-mapping study of the murine monocyte and macrophage compartment taking advantage of constitutive and conditional CX(3)CR1 promoter-driven Cre recombinase expression is reported, establishing that short-lived Ly6C(+) monocytes constitute obligatory steady-state precursors of blood-resident Ly 6C(-) cells and that the abundance of Ly6 C(+) blood monocytes dynamically controls the circulation lifespan of their progeny.

TL;DR: Using transcriptomic profiling of flow-sorted cells, it is found that monocyte to alveolar macrophage differentiation unfolds continuously over the course of fibrosis and its resolution, and suggests that selectively targeting alveolars macrophages differentiation within the lung may ameliorate fibrosis without the adverse consequences associated with global monocyte or tissue-resident alve Polar Macrophage depletion.
