Profiling early head and neck cancer.
TLDR
Prevention and early diagnosis of high-risk pre-malignant lesions are high priorities for reducing deaths due to head and neck cancer.Abstract:
Head and neck squamous-cell carcinoma (HNSCC) is the sixth most common cancer worldwide and, disappointingly, survival rates are not improving. Moreover, HNSCC has a severe impact on the quality of life of patients and survivors, and the significant morbidity subsequent to treatment often mandates long-term multidisciplinary care, which places significant financial pressures on the treating institution. Therefore, prevention and early diagnosis of high-risk pre-malignant lesions are high priorities for reducing deaths due to head and neck cancer. Recent advances have begun to elucidate the different aetiologies of HNSCCs in relation to previous pre-malignancies and to identify which pre-malignant lesions are likely to progress to malignancy.read more

S100 proteins in cancer
TL;DR: Emerging in vivo evidence indicates that the biology of most S100 proteins is complex and multifactorial, and that these proteins actively contribute to tumorigenic processes such as cell proliferation, metastasis, angiogenesis and immune evasion.

TL;DR: Findings in primary human tumors provide novel biomarkers for early detection of HPV(+) and HPV(-) cancers, and emphasize the potential value of targeting E6 and E7 function, alone or combined with radiation and/or traditional chemotherapy, in the treatment of HPV (+) cancers.

NF-κB addiction and its role in cancer: ‘one size does not fit all’
TL;DR: Multiple mechanisms of NF-κB activation and their regulation by multitargeted agent in contrast to monotargeted agents are discussed, thus ‘one size does not fit all’ cancers.

TL;DR: The emerging information on how the use of modern technologies and the molecular dissection of aberrant signaling networks, including the EGFR, ras, NFkappaB, Stat, Wnt/beta-catenin, TGF-beta, and PI3K-AKT-mTOR signaling pathways can help elucidate the molecular mechanisms underlying HNSCC progression is discussed.

Optimal treatment for recurrent/metastatic head and neck cancer
Jan B. Vermorken,Pol Specenier +1 more
TL;DR: A recent European randomized trial showed that adding cetuximab, the first clinically available EGFR-directed monoclonal antibody, to a standard chemotherapy regimen (platinum/5-fluorouracil) leads to an important survival benefit and this, with support of an additional smaller study in the US, has changed practice.

Head and Neck Cancer Incidence Trends in Young Americans, 1973-1997, With a Special Analysis for Tongue Cancer
Stimson P. Schantz,Guo-Pei Yu +1 more
TL;DR: A sharp increasing trend in tongue cancer in young Americans may be attributed to persons born after 1938, which suggests the emergence of a distinct disease process that is apparent in white but not black populations.

Distinct pattern of expression of differentiation and growth-related genes in squamous cell carcinomas of the head and neck revealed by the use of laser capture microdissection and cDNA arrays.

TL;DR: The successful use of laser capture microdissection to procure specific cell populations is shown, and it is found that most HNSCC overexpress members of the wnt and notch growth and differentiation regulatory system, thus suggesting that the wNT and notch pathways may contribute in squamous cell carcinogenesis.

TL;DR: A novel classification of the secondary tumors that develop after treatment of a carcinoma in the oral cavity or oropharynx is described, and definitions for a “true SPT,” a local recurrence, a ”SFT” (second field tumor derived from the same genetically altered mucosal field as the primary tumor), and a metastasis are proposed.
