This work has defined the 13 bp palindrome GGTCACAGTGACC as a minimal functional estrogen responsive element (ERE), which binds estrogen receptor preferentially in vitro and point mutations within the ERE decrease its affinity for the estrogen receptor and result in a complete loss of estrogen inducibility.Abstract:
Sequences located upstream of the transcription initiation site of the Xenopus vitellogenin A2 (vit A2) gene contain a hormone dependent enhancer that confers estrogen control to the heterologous thymidine kinase (tk) promoter. As a minimal functional estrogen responsive element (ERE), we have defined the 13 bp palindrome GGTCACAGTGACC. This ERE binds estrogen receptor preferentially in vitro. Although the ERE shares some structural features with the glucocorticoid responsive element (GRE) it is distinct from this element since it neither binds glucocorticoid receptor in vitro nor does it confer glucocorticoid inducibility to a fusion gene. Point mutations within the ERE decrease its affinity for the estrogen receptor and result in a complete loss of estrogen inducibility.read more

TL;DR: A new dimension to the p53 master regulatory network is established where p53-mediated transcription from a ½ site RE can be determined by ER binding at one or more cis-acting EREs in manner that is dependent on level of ER protein, the type of ER ligand and the specific p 53-inducing agent.

Oestrogen receptor-alpha regulates non-canonical Hedgehog-signalling in the mammary gland.
TL;DR: It is demonstrated here that conditional expression of ERα in luminal mammary epithelial cells on the mes background facilitates ductal morphogenesis with kinetics similar to that of the MMTV-c-src(Act) mice.

TL;DR: The results suggest that ER is able to bind to ERE independently of the presence of estradiol or other proteins and, therefore, that est radiol does not act by modulating the ability of ER to bindto ERE on DNA.

Identification of two functional estrogen response elements complexed with AP-1-like sites in the human insulin receptor gene promoter.
TL;DR: These data provide the first identification of EREs complexed with AP-1-like sites in the hIR promoter, which account for the transcriptional repression of the hir gene mediated by ERbeta in U-937 cells.

TL;DR: Over-expression of the Xenopus estrogen receptor in a vaccinia virus-derived expression system revealed that the receptor localized exclusively in the nucleus of the infected cells, irrespective of the presence or absence of the ligand.
