Overview and findings from the religious orders study
TLDR
This manuscript summarizes the study methods including the study design and describes the clinical evaluation, assessment of risk factors, collection of ante-mortem biological specimens, brain autopsy and collection of selected postmortem data.Abstract:
The Religious Orders Study is a longitudinal clinical-pathologic cohort study of aging and Alzheimer’s disease (AD). In this manuscript, we summarize the study methods including the study design and describe the clinical evaluation, assessment of risk factors, collection of ante-mortem biological specimens, brain autopsy and collection of selected post-mortem data. The results: 1) review the relation of neuropathologic indices to clinical diagnoses and cognition proximate to death; 2) examine the relation of risk factors to clinical outcomes; 3) examine the relation of risk factors to measures of neuropathology; and 4) summarize additional study findings. We then discuss and contextualize the study findings.read more

TL;DR: A new epigenetic biomarker of aging, DNAm PhenoAge, is developed that strongly outperforms previous measures in regards to predictions for a variety of aging outcomes, including all-cause mortality, cancers, healthspan, physical functioning, and Alzheimer's disease.

TL;DR: Gingipains from Porphyromonas gingivalis drive Alzheimer’s pathology and can be blocked with small-molecule inhibitors, suggesting that gingipain inhibitors could be valuable for treating P. gedivalis brain colonization and neurodegeneration in Alzheimer's disease.

Bernstein,Alexander Meissner,Nilufer Ertekin-Taner,Lori B. Chibnik,Manolis Kellis,Jonathan Mill,David A. Bennett +28 more
TL;DR: The analyses suggest that DNA methylation changes may have a role in the onset of AD given that they were observed in presymptomatic subjects and that six of the validated genes connect to a known AD susceptibility gene network.

REST and stress resistance in ageing and Alzheimer’s disease

TL;DR: It is shown that induction of the repressor element 1-silencing transcription factor (REST) is a universal feature of normal ageing in human cortical and hippocampal neurons, and levels during ageing are closely correlated with cognitive preservation and longevity.

A practical method for grading the cognitive state of patients for the clinician
TL;DR: The Mini-Mental State (MMS) as mentioned in this paper is a simplified version of the standard WAIS with eleven questions and requires only 5-10 min to administer, and is therefore practical to use serially and routinely.
