Abstract: Heterozygous mutations in GBA1, the gene encoding lysosomal glucocerebrosidase, are the most frequent known genetic risk factor for Parkinson’s disease. Reduced glucocerebrosidase and α-synuclein accumulation are directly related in cell models of Parkinson’s disease. We investigated relationships between Parkinson’s disease-specific glucocerebrosidase deficits, glucocerebrosidase-related pathways, and α-synuclein levels in brain tissue from subjects with sporadic Parkinson’s disease without GBA1 mutations. Brain regions with and without a Parkinson’s disease-related increase in α-synuclein levels were assessed in autopsy samples from subjects with sporadic Parkinson’s disease (n = 19) and age- and post-mortem delay-matched controls (n = 10). Levels of glucocerebrosidase, α-synuclein and related lysosomal and autophagic proteins were assessed by western blotting. Glucocerebrosidase enzyme activity was measured using a fluorimetric assay, and glucocerebrosidase and α-synuclein messenger RNA expression determined by quantitative polymerase chain reaction. Related sphingolipids were analysed by mass spectrometry. Multivariate statistical analyses were performed to identify differences between disease groups and regions, with non-parametric correlations used to identify relationships between variables. Glucocerebrosidase protein levels and enzyme activity were selectively reduced in the early stages of Parkinson’s disease in regions with increased α-synuclein levels although limited inclusion formation, whereas GBA1 messenger RNA expression was non-selectively reduced in Parkinson’s disease. The selective loss of lysosomal glucocerebrosidase was directly related to reduced lysosomal chaperone-mediated autophagy, increased α-synuclein and decreased ceramide. Glucocerebrosidase deficits in sporadic Parkinson’s disease are related to the abnormal accumulation of α-synuclein and are associated with substantial alterations in lysosomal chaperone-mediated autophagy pathways and lipid metabolism. Our data suggest that the early selective Parkinson’s disease changes are likely a result of the redistribution of cellular membrane proteins leading to a chronic reduction in lysosome function in brain regions vulnerable to Parkinson’s disease pathology.

Abstract: flexmix provides infrastructure for flexible fitting of finite mixture models in R using the expectation-maximization (EM) algorithm or one of its variants. The functionality of the package was enhanced. Now concomitant variable models as well as varying and constant parameters for the component specific generalized linear regression models can be fitted. The application of the package is demonstrated on several examples, the implementation described and examples given to illustrate how new drivers for the component specific models and the concomitant variable models can be defined.

Abstract: The extraordinary properties of graphene and carbon nanotubes motivate the development of methods for their use in producing continuous, strong, tough fibres. Previous work has shown that the toughness of the carbon nanotube-reinforced polymer fibres exceeds that of previously known materials. Here we show that further increased toughness results from combining carbon nanotubes and reduced graphene oxide flakes in solution-spun polymer fibres. The gravimetric toughness approaches 1,000 J g(-1), far exceeding spider dragline silk (165 J g(-1)) and Kevlar (78 J g(-1)). This toughness enhancement is consistent with the observed formation of an interconnected network of partially aligned reduced graphene oxide flakes and carbon nanotubes during solution spinning, which act to deflect cracks and allow energy-consuming polymer deformation. Toughness is sensitive to the volume ratio of the reduced graphene oxide flakes to the carbon nanotubes in the spinning solution and the degree of graphene oxidation. The hybrid fibres were sewable and weavable, and could be shaped into high-modulus helical springs.

Abstract: Potassium-ion batteries (PIBs) are a promising alternative to lithium-ion batteries because potassium is an abundant natural resource. To date, PIBs are in the early stages of exploration and only a few anode materials have been investigated. This study reports a cobalt sulfide and graphene (CoS@G) composite as anode electrode for PIBs for the first time. The composite features interconnect quantum dots of CoS nanoclusters uniformly anchored on graphene nanosheets. The coexistence of CoS quantum dot nanoclusters and graphene nanosheets endows the composite with large surface area, highly conductive network, robust structural stability, and excellent electrochemical energy storage performance. An unprecedented capacity of 310.8 mA h g−1 at 500 mA g−1 is obtained after 100 cycles, with a rate capability better than an equivalent sodium-ion batteries (SIBs). This work provides the evidence that PIBs can be a promising alternative to SIBs, especially at high charge–discharge rates. The development of the CoS@G anode material also provides the basis of expanding the library of suitable anode materials for PIBs.

Abstract: Although the term “behavioral epidemiology” has been used in the literature since the late 1970s, it has not been clearly defined. A behavioral epidemiology framework is proposed to specify a systematic sequence of studies on health-related behaviors, leading to evidence-based interventions directed at populations. The phases are: 1—establish links between behaviors and health; 2—develop measures of the behavior; 3—identify influences on the behavior; 4—evaluate interventions to change the behavior; 5—translate research into practice. Mature research areas are expected to have more studies in the latter phases. Recent volumes of four journals (Annals of Behavioral Medicine, Health Psychology, Journal of Nutrition Education, Tobacco Control) were audited, and empirical studies were classified into these phases. Phase 3 studies were common (identifying influences on behaviors; 27% to 50%), and Phase 2 studies were least common (measurement; 0% to 15%).Annals of Behavioral Medicine andHealth Psychology were low on Phase 4 (intervention studies; 9% and 11%, respectively). TheJournal of Nutrition Education was the only journal reviewed that had a substantial number (20%) of Phase 5 studies (translating research into practice). The behavioral epidemiology framework can be used to evaluate the status of research on health behaviors and to guide research policies.
