Abstract: We studied the formation of the Himalayan mountain range and the Tibetan Plateau by investigating their lithospheric structure. Using an 800-kilometer-long, densely spaced seismic array, we have constructed an image of the crust and upper mantle beneath the Himalayas and the southern Tibetan Plateau. The image reveals in a continuous fashion the Main Himalayan thrust fault as it extends from a shallow depth under Nepal to the mid-crust under southern Tibet. Indian crust can be traced to 31°N. The crust/mantle interface beneath Tibet is anisotropic, indicating shearing during its formation. The dipping mantle fabric suggests that the Indian mantle is subducting in a diffuse fashion along several evolving subparallel structures.

Abstract: Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) are life-threatening adverse drug reactions characterized by massive epidermal necrosis, in which the specific danger signals involved remain unclear. Here we show that blister cells from skin lesions of SJS-TEN primarily consist of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, and both blister fluids and cells were cytotoxic. Gene expression profiling identified granulysin as the most highly expressed cytotoxic molecule, confirmed by quantitative PCR and immunohistochemistry. Granulysin concentrations in the blister fluids were two to four orders of magnitude higher than perforin, granzyme B or soluble Fas ligand concentrations, and depleting granulysin reduced the cytotoxicity. Granulysin in the blister fluids was a 15-kDa secretory form, and injection of it into mouse skin resulted in features mimicking SJS-TEN. Our findings demonstrate that secretory granulysin is a key molecule responsible for the disseminated keratinocyte death in SJS-TEN and highlight a mechanism for CTL- or NK cell—mediated cytotoxicity that does not require direct cellular contact. Abstract: Security remains a major roadblock to universal acceptance of the Web for many kinds of transactions, especially since the recent sharp increase in remotely exploitable vulnerabilities have been attributed to Web application bugs. Many verification tools are discovering previously unknown vulnerabilities in legacy C programs, raising hopes that the same success can be achieved with Web applications. In this paper, we describe a sound and holistic approach to ensuring Web application security. Viewing Web application vulnerabilities as a secure information flow problem, we created a lattice-based static analysis algorithm derived from type systems and typestate, and addressed its soundness. During the analysis, sections of code considered vulnerable are instrumented with runtime guards, thus securing Web applications in the absence of user intervention. With sufficient annotations, runtime overhead can be reduced to zero. We also created a tool named.WebSSARI (Web application Security by Static Analysis and Runtime Inspection) to test our algorithm, and used it to verify 230 open-source Web application projects on SourceForge.net, which were selected to represent projects of different maturity, popularity, and scale. 69 contained vulnerabilities. After notifying the developers, 38 acknowledged our findings and stated their plans to provide patches. Our statistics also show that static analysis reduced potential runtime overhead by 98.4%.

Abstract: Recent studies have demonstrated the important role of plant microRNAs (miRNAs) under nutrient deficiencies. In this study, deep sequencing of Arabidopsis (Arabidopsis thaliana) small RNAs was conducted to reveal miRNAs and other small RNAs that were differentially expressed in response to phosphate (Pi) deficiency. About 3.5 million sequence reads corresponding to 0.6 to 1.2 million unique sequence tags from each Pi-sufficient or Pi-deficient root or shoot sample were mapped to the Arabidopsis genome. We showed that upon Pi deprivation, the expression of miR156, miR399, miR778, miR827, and miR2111 was induced, whereas the expression of miR169, miR395, and miR398 was repressed. We found cross talk coordinated by these miRNAs under different nutrient deficiencies. In addition to miRNAs, we identified one Pi starvation-induced DICER-LIKE1-dependent small RNA derived from the long terminal repeat of a retrotransposon and a group of 19-nucleotide small RNAs corresponding to the 5' end of tRNA and expressed at a high level in Pi-starved roots. Importantly, we observed an increased abundance of TAS4-derived trans-acting small interfering RNAs (ta-siRNAs) in Pi-deficient shoots and uncovered an autoregulatory mechanism of PAP1/MYB75 via miR828 and TAS4-siR81(-) that regulates the biosynthesis of anthocyanin. This finding sheds light on the regulatory network between miRNA/ta-siRNA and its target gene. Of note, a substantial amount of miR399* accumulated under Pi deficiency. Like miR399, miR399* can move across the graft junction, implying a potential biological role for miR399*. This study represents a comprehensive expression profiling of Pi-responsive small RNAs and advances our understanding of the regulation of Pi homeostasis mediated by small RNAs.
