Abstract: Monitoring the behavior of single molecules in living cells is a powerful approach to investigate the details of cellular processes. Owing to their optical, chemical and biofunctional properties, semiconductor quantum dot (QD) probes promise to be tools of choice in this endeavor. Here we review recent advances that allow ever more controlled experiments at the single-nanoparticle level in live cells. Several examples, related to membrane dynamics, cell signaling or intracellular transport, illustrate how single QD tracking can be readily used to decipher complex biological processes and address key concepts that underlie cellular organization and dynamics.

Abstract: With advances in knowledge disease, boundaries may change. Occasionally, these changes are of such a magnitude that they require redefinition of the disease. In recognition of the profound changes in our understanding of Parkinson's disease (PD), the International Parkinson and Movement Disorders Society (MDS) commissioned a task force to consider a redefinition of PD. This review is a discussion article, intended as the introductory statement of the task force. Several critical issues were identified that challenge current PD definitions. First, new findings challenge the central role of the classical pathologic criteria as the arbiter of diagnosis, notably genetic cases without synuclein deposition, the high prevalence of incidental Lewy body (LB) deposition, and the nonmotor prodrome of PD. It remains unclear, however, whether these challenges merit a change in the pathologic gold standard, especially considering the limitations of alternate gold standards. Second, the increasing recognition of dementia in PD challenges the distinction between diffuse LB disease and PD. Consideration might be given to removing dementia as an exclusion criterion for PD diagnosis. Third, there is increasing recognition of disease heterogeneity, suggesting that PD subtypes should be formally identified; however, current subtype classifications may not be sufficiently robust to warrant formal delineation. Fourth, the recognition of a nonmotor prodrome of PD requires that new diagnostic criteria for early-stage and prodromal PD should be created; here, essential features of these criteria are proposed. Finally, there is a need to create new MDS diagnostic criteria that take these changes in disease definition into consideration.

Abstract: Some general formulae are obtained for size-biased sampling from a Poisson point process in an abstract space where the size of a point is defined by an arbitrary strictly positive function. These formulae explain why in certain cases (gamma and stable) the size-biased permutation of the normalized jumps of a subordinator can be represented by a stickbreaking (residual allocation) scheme defined by independent beta random variables. An application is made to length biased sampling of excursions of a Markov process away from a recurrent point of its statespace, with emphasis on the Brownian and Bessel cases when the associated inverse local time is a stable subordinator. Results in this case are linked to generalizations of the arcsine law for the fraction of time spent positive by Brownian motion.

Abstract: Climate feedback analysis constitutes a useful framework for comparing the global mean surface temperature responses to an external forcing predicted by general circulation models (GCMs). Nevertheless, the contributions of the different radiative feedbacks to global warming (in equilibrium or transient conditions) and their comparison with the contribution of other processes (e.g., the ocean heat uptake) have not been quantified explicitly. Here these contributions from the classical feedback analysis framework are defined and quantified for an ensemble of 12 third phase of the Coupled Model Intercomparison Project (CMIP3)/Intergovernmental Panel on Climate Change (IPCC) Fourth Assessment Report (AR4) coupled atmosphere–ocean GCMs. In transient simulations, the multimodel mean contributions to global warming associated with the combined water vapor–lapse-rate feedback, cloud feedback, and ocean heat uptake are comparable.
