Abstract: In order to identify the neutralizing epitope of the porcine epidemic diarrhea virus (PEDV), the spike protein region that is presumed to contain the virus-neutralizing epitope was determined This was based on the sequence information for the neutralizing epitope of the transmissible gastroenteritis virus (TGEV) A recombinant protein that corresponds to the spike region of TGEV was produced, and polyclonal antisera were generated using the recombinant protein It was discovered that polyclonal antisera significantly inhibited plaque formation by PEDV, suggesting that this region of the spike protein contains the epitope(s) that is capable of inducing PEDV-neutralizing antibodies In addition, the region that corresponds to the neutralizing epitope of TGEV may also be involved in neutralizing PEDV, although the two viruses are serologically quite distinct Finally, the amino acid sequences that are deduced from the genes for the determined-neutralizing epitope were highly homologous among the PEDV strains that were isolated from different geographical areas, which suggests conservation of the antigen gene

Abstract: Alumina-borate/PVA composite fibres were prepared using sol–gel processing and an electrospinning technique. After calcination of the thin fibres, ultra-fine fibres of alumina-borate oxide with a diameter of about 550 nm could be prepared. The fibres were characterized by SEM, XRD and FT-IR. The results showed that the crystalline phase and morphology of alumina-borate fibres were largely influenced by the calcination temperature.

Abstract: The normalized differential cross section for top quark pair (tt) production is measured in pp collisions at a centre-of-mass energy of 8TeV at the CERN LHC using the CMS detector in data corresponding to an integrated luminosity of 19.7fb^(−1). The measurements are performed in the lepton+jets (e/μ +jets) and in the dilepton (e^+e^−, μ^+μ^−, and e^±μ^∓) decay channels. The tt cross section is measured as a function of the kinematic properties of the charged leptons, the jets associated to b quarks, the top quarks, and the tt system. The data are compared with several predictions from perturbative quantum chromodynamic up to approximate next-to-next-to-leading-order precision. No significant deviations are observed relative to the standard model predictions.

Abstract: Doxorubicin (DOX) is one of the most potent anticancer drugs and induces acute cardiac arrhythmias and chronic cumulative cardiomyopathy. Though DOX-induced cardiotoxicity is known to be caused mainly by ROS generation, a disturbance of Ca2+ homeostasis is also implicated one of the cardiotoxic mechanisms. In this study, a molecular basis of DOX-induced modulation of intracellular Ca2+ concentration ([Ca2+]i) was investigated. Treatment of adult rat cardiomyocytes with DOX increased [Ca2+]i irrespectively of extracellular Ca2+, indicating DOX-mediated Ca2+ release from intracellular Ca2+ stores. The DOX-induced Ca2+ increase was slowly processed and sustained. The Ca2+ increase was inhibited by pretreatment with a sarcoplasmic reticulum (SR) Ca2+ channel blocker, ryanodine or dantrolene, and an antioxidant, alpha-lipoic acid or alpha-tocopherol. DOX-induced ROS generation was observed immediately after DOX treatment and increased in a time-dependent manner. The ROS production was significantly reduced by the pretreatment of the SR Ca2+ channel blockers and the antioxidants. Moreover, DOX-mediated activation of caspase-3 was significantly inhibited by the Ca2+ channel blockers and a-lipoic acid but not a-tocopherol. In addition, cotreatment of ryanodine with alpha-lipoic acid resulted in further inhibition of the casapse-3 activity. These results demonstrate that DOX-mediated ROS opens ryanodine receptor, resulting in an increase in [Ca2+]i and that the increased [Ca2+]i induces ROS production. These observations also suggest that DOX/ROS-induced increase of [Ca2+]i plays a critical role in damage of cardiomyocytes.
