Memory B Cells Activate Brain-Homing, Autoreactive CD4+ T Cells in Multiple Sclerosis.

TLDR
It is demonstrated that self-reactivity, defined as “autoproliferation” of peripheral Th1 cells, is elevated in patients carrying the HLA-DR15 haplotype, and RASGRP2 is identified as target autoantigen that is expressed in the brain and B cells.

B Cell Responses: Cell Interaction Dynamics and Decisions

TL;DR: How basic research is informing efforts to generate vaccines that induce broadly neutralizing antibodies against viral pathogens is discussed, revealing the special features associated with allergen-reactive IgE responses and uncovering the antibody-independent mechanisms by which B cells contribute to health and disease.
Journal ArticleDOI

B cell depletion therapies in autoimmune disease: advances and mechanistic insights

TL;DR: The understanding of B cell biology is updated, the results of clinical trials using BCDT in autoimmune indications are reviewed, hypotheses for the mechanism of action are discussed, and evolving strategies for targeting B cells beyond depletion are speculated on.

Conventional DCs sample and present myelin antigens in the healthy CNS and allow parenchymal T cell entry to initiate neuroinflammation

TL;DR: A population of conventional dendritic cells, but not border-associated macrophages or microglia, are found to be essential for licensing T cells to initiate neuroinflammation.

edgeR: a Bioconductor package for differential expression analysis of digital gene expression data.

TL;DR: EdgeR as mentioned in this paper is a Bioconductor software package for examining differential expression of replicated count data, which uses an overdispersed Poisson model to account for both biological and technical variability and empirical Bayes methods are used to moderate the degree of overdispersion across transcripts, improving the reliability of inference.

RSEM: accurate transcript quantification from RNA-Seq data with or without a reference genome

TL;DR: It is shown that accurate gene-level abundance estimates are best obtained with large numbers of short single-end reads, and estimates of the relative frequencies of isoforms within single genes may be improved through the use of paired- end reads, depending on the number of possible splice forms for each gene.
Journal ArticleDOI

Universal sample preparation method for proteome analysis

TL;DR: A method is described, filter-aided sample preparation (FASP), which combines the advantages of in-gel and in-solution digestion for mass spectrometry–based proteomics and allows single-run analyses of organelles and an unprecedented depth of proteome coverage.

TL;DR: In this article, a collaborative GWAS involving 9,772 cases of European descent collected by 23 research groups working in 15 different countries, they have replicated almost all of the previously suggested associations and identified at least a further 29 novel susceptibility loci.
