Journal ArticleDOI

Limitations of Constant-Force-Feedback Experiments

TL;DR: It is demonstrated that the commonly used technique of force feedback has severe limitations when used to evaluate rapid macromolecular conformational transitions, and the causes are elucidated and a simple test is provided to identify and evaluate the magnitude of the effect.
Journal ArticleDOI

Real-Time Imaging of the Reorientation Mechanisms of YOYO-Labelled DNA Molecules during 90° and 120° Pulsed Field Gel Electrophoresis

TL;DR: The first clear observations of individual molecules of DNA during the reorientation process in 90 degrees PFGE and 120 degreesPFGE are shown and computer simulations used to reproduce the molecular mechanisms of motion as well as the DNA separation features are discussed.
Journal ArticleDOI

Interaction of water‐soluble porphyrins with single‐ and double‐stranded polyribonucleotides

TL;DR: CD and uv‐visible absorption studies with several tetracationic water‐soluble porphyrin derivatives show that some of these species can serve as probes to discriminate between A‐ and B‐conformational forms of single‐stranded polynucleotides.
Journal ArticleDOI

Trigger loop folding determines transcription rate of Escherichia coli’s RNA polymerase

TL;DR: Single molecule experiments with the WT and two TL-tip mutants of the Escherichia coli enzyme reveal that tip mutations modulate RNAP’s pause-free velocity, identifying TL conformational changes as one of two rate-determining steps in elongation.
