Rationally designed BCL6 inhibitors target activated B cell diffuse large B cell lymphoma
Mariano G. Cardenas,Wenbo Yu,Wendy Béguelin,Matthew R. Teater,Huimin Geng,Huimin Geng,Rebecca Goldstein,Erin Oswald,Katerina Hatzi,Shao Ning Yang,Joanna Cohen,Rita Shaknovich,Kenno Vanommeslaeghe,Kenno Vanommeslaeghe,Huimin Cheng,Dongdong Liang,Hyo Je Cho,Joshua Abbott,Wayne Tam,Wei Du,John P. Leonard,Olivier Elemento,Leandro Cerchietti,Tomasz Cierpicki,Fengtian Xue,Alexander D. MacKerell,Ari Melnick +26 more
TL;DR: Small-molecule inhibitors that display higher affinity for BCL6 than its endogenous corepressor ligands are designed to evaluate their therapeutic efficacy for targeting ABC-DLBCL and indicate that ABC- DLBCL is a BCL 6-dependent disease that can be targeted by rationally designed inhibitors that exceed the binding affinity of natural BCL7 ligands.
Journal ArticleDOI
Mechanosensitive pannexin-1 channels mediate microvascular metastatic cell survival
Furlow Pw,Sicong Zhang,Soong Td,Nils Halberg,Hani Goodarzi,Mangrum C,Wu Yg,Olivier Elemento,Sohail F. Tavazoie +8 more
TL;DR: A mutation encoding a truncated form of the pannexin-1 (PANX1) channel, PANX11–89, is identified as recurrently enriched in highly metastatic breast cancer cells, suggesting a molecular basis for metastatic cell survival on microvasculature-induced biomechanical trauma.
Journal ArticleDOI
Widespread Mitotic Bookmarking by Histone Marks and Transcription Factors in Pluripotent Stem Cells
Yiyuan Liu,Bobbie Pelham-Webb,Bobbie Pelham-Webb,Dafne Campigli Di Giammartino,Jiexi Li,Daleum Kim,Katsuhiro Kita,Néstor Saiz,Vidur Garg,Ashley S. Doane,Paraskevi Giannakakou,Anna-Katerina Hadjantonakis,Olivier Elemento,Effie Apostolou +13 more
TL;DR: It is shown that, in pluripotent stem cells, certain histone marks and stem cell regulators remain associated with specific genomic regions of mitotic chromatin, a phenomenon known as mitotic bookmarking.
Journal ArticleDOI
Cancer systems biology: embracing complexity to develop better anticancer therapeutic strategies
Wei Du,Olivier Elemento +1 more
TL;DR: Implementing patient-specific computational and mathematical models of cancer will significantly improve the specificity and efficacy of targeted therapy, and will accelerate the adoption of personalized and precision cancer medicine.
Journal ArticleDOI
Deficiency of UBE2T, the E2 Ubiquitin Ligase Necessary for FANCD2 and FANCI Ubiquitination, Causes FA-T Subtype of Fanconi Anemia.
Kimberly A. Rickman,Francis P. Lach,Avinash Abhyankar,Frank X. Donovan,Erica Sanborn,Jennifer A. Kennedy,Carrie Sougnez,Stacey Gabriel,Olivier Elemento,Settara C. Chandrasekharappa,Detlev Schindler,Arleen D. Auerbach,Agata Smogorzewska +12 more
TL;DR: An individual presenting with typical FA features who is deficient for the ubiquitin-conjugating enzyme (E2), UBE2T is described, demonstrating that deficiency of the protein UBE1T can lead to Fanconi anemia.
