Elevated HLA-E and NKG2A as a Consequence of Chronic Immune Activation Defines Resistance to M. bovis BCG Immunotherapy in Non-Muscle-Invasive Bladder Cancer
Daniel Ranti,Y.A. Wang,Jorge Daza,Carolyn Bieber,Bérengère Salomé,Elliot Merritt,Julie-Ann Cavallo,E. Hegewisch-Solloa,Emily M. Mace,Adam M. Farkas,S. Shroff,Foramben Patel,Michelle Fang Tran,Trine Strandgaard,Sia Viborg Lindskrog,Lars Dyrskjøt,Jingjing Qi,M. Patel,Daniel Geanon,Geoffrey Kelly,R. M. de Real,Bunsho Lee,Seunghee Kim-Schulze,Tin Htwe Thin,Monica Garcia-Barros,Kristin G. Beaumont,Hiranmayi Ravichandran,Y Hu,Y. Wang,L. Wang,Dominic LaRoche,Y K Lee,Robert Sebra,Rachel I. Brody,Olivier Elemento,Anna S. Tocheva,Brooks Hopkins,Peter Wiklund,J. G. Zhu,Matthew D. Galsky,Nina Bhardwaj,John P. Sfakianos,Amir Horowitz +42 more
TL;DR: Combined NKG2A and PD-L1 blockade for BCG unresponsive disease is supported, and in situ spatial analyses revealed that HLA-EHIGH tumors are activated to recruit NK and T cells via chemokine production, potentially sparing Hla-ELOW tumors that would otherwise be susceptible to lysis.
Journal ArticleDOI
Single-cell RNA sequencing reveals immunosuppressive myeloid cell diversity during malignant progression in a murine model of glioma.
Sakthi Rajendran,Yang Hu,Alessandro Canella,Clayton R. Peterson,Amy C. Gross,Maren Cam,Matthew Nazzaro,Abigail Haffey,Akdes Serin-Harmanci,Rosario Distefano,Giovanni Nigita,Wesley Wang,Daniel Kreatsoulas,Zihai Li,Jesse A. Sepeda,Andrew Sas,Mark E. Hester,Katherine E. Miller,Olivier Elemento,Ryan D. Roberts,Eric C. Holland,Ganesh Rao,Elaine R. Mardis,Prajwal Rajappa +23 more
TL;DR: In this article , the authors investigate the cellular heterogeneity of the TME using single-cell RNA sequencing in a murine glioma model that recapitulates the malignant progression of LGG to HGG.

Abstract A31: ERG-mediated alterations in chromatin conformation
David S. Rickman,Ari Melnick,Olivier Elemento,Mark A. Rubin,David Soong,Benjamin J. Moss,Juan Miguel Mosquera,Jan Dlabal,Stéphane Terry,Theresa Y. MacDonald,Karen L. Bunting,Francesca Demichelis +11 more
TL;DR: It is demonstrated for the first time that the oncogenic transcription factor ERG can induce global, reproducible and functionally coherent changes in chromatin organization that are associated with gene expression differences and genomic alterations.

Abstract 134: Immunogenomic landscape of neuroendocrine prostate cancer (NEPC)
Alison Ferguson,Bhavneet Bhinder,Vincenza Conteduca,Michael Sigouros,Andrea Sboner,David M. Nanus,Scott T. Tagawa,David S. Rickman,Olivier Elemento,Himisha Beltran +9 more
TL;DR: Fergusson et al. as discussed by the authors investigated the immune landscape of NEPC in relation to prostate cancer subtypes and SCLC to identify potential targets, and found that NEPC is characterized by a relatively ‘cold' tumor immune microenvironment similar to other metastatic prostate cancer, but higher PDL1 expression comparable to small cell lung carcinoma.
