Journal ArticleDOI
Inherited determinants of early recurrent somatic mutations in prostate cancer.
Alessandro Romanel,Sonia Garritano,Blerta Stringa,Mirjam Blattner,Davide Dalfovo,Dimple Chakravarty,David Soong,Kellie A. Cotter,Gianluca Petris,Priyanka Dhingra,Paola Gasperini,Anna Cereseto,Olivier Elemento,Olivier Elemento,Andrea Sboner,Ekta Khurana,Ekta Khurana,Alberto Inga,Mark A. Rubin,Mark A. Rubin,Francesca Demichelis,Francesca Demichelis +21 more
TL;DR: A ranking score and benign prostate transcriptomes are used to identify a non-coding polymorphic regulatory element at 7p14.3 that associates with DNA repair and hormone-regulated transcript levels and with an early recurrent prostate cancer-specific somatic mutation in the Speckle-Type POZ protein (SPOP) gene.

A harmonized meta-knowledgebase of clinical interpretations of cancer genomic variants
Alex H. Wagner,Brian Walsh,Georgia Mayfield,David Tamborero,David Tamborero,Dmitriy Sonkin,Kilannin Krysiak,Jordi Deu-Pons,Ryan P Duren,Jianjiong Gao,Julie A. McMurry,Sara E. Patterson,Catherine Del Vecchio Fitz,Ozman Ugur Sezerman,Jeremy L. Warner,Damian T. Rieke,Tero Aittokallio,Ethan Cerami,Deborah I. Ritter,Lynn M. Schriml,Melissa A. Haendel,Gordana Raca,Subha Madhavan,Michael Baudis,Jacques S. Beckmann,Rodrigo Dienstmann,Debyani Chakravarty,Xuan Shirley Li,Susan M. Mockus,Olivier Elemento,Nikolaus Schultz,Nuria Lopez-Bigas,Nuria Lopez-Bigas,Mark Lawler,Jeremy Goecks,Malachi Griffith,Obi L. Griffith,Adam A. Margolin +37 more
TL;DR: A framework for aggregating and harmonizing variant interpretations is developed to produce a meta-knowledgebase of 12,856 aggregate interpretations covering 3,437 unique variants in 415 genes, 357 diseases, and 791 drugs, illuminating the need for open, interoperable sharing of variant interpretation data.
Journal ArticleDOI
Therapeutic efficacy of the bromodomain inhibitor OTX015/MK-8628 in ALK-positive anaplastic large cell lymphoma: an alternative modality to overcome resistant phenotypes
Michela Boi,Michela Boi,Maria Todaro,Maria Todaro,Valentina Vurchio,Shao Ning Yang,John Moon,Ivo Kwee,Ivo Kwee,Andrea Rinaldi,Heng Pan,Ramona Crescenzo,Ramona Crescenzo,Mangeng Cheng,Leandro Cerchietti,Olivier Elemento,Maria E. Riveiro,Esteban Cvitkovic,Francesco Bertoni,Giorgio Inghirami,Giorgio Inghirami,Giorgio Inghirami +21 more
TL;DR: OtX015 clinical trials in refractory ALCL in combination with inhibitors of interleukin-2-inducible kinase or SHH/GLI1 are supported, and a novel bromodomain inhibitor is investigated in ALK-positive ALCLs.
Journal ArticleDOI
Akt Suppression of TGFβ Signaling Contributes to the Maintenance of Vascular Identity in Embryonic Stem Cell-Derived Endothelial Cells
Edo Israely,Michael Ginsberg,Daniel J. Nolan,Bi-Sen Ding,Daylon James,Olivier Elemento,Shahin Rafii,Sina Y. Rabbany +7 more
TL;DR: Findings identify a role for Akt in regulating the developmental potential of ES cell‐derived ECs and demonstrate that active Akt maintains endothelial identity in embryonic ECs by interfering with active TGFβ‐mediated processes that would ordinarily usher these cells to alternate fates.
Journal ArticleDOI
Identification of Reprogrammed Myeloid Cell Transcriptomes in NSCLC.
Anna Durrans,Dingcheng Gao,Ravi Gupta,Kari R. Fischer,Hyejin Choi,Tina El Rayes,Seongho Ryu,Abu Nasar,Cathy F. Spinelli,Weston Andrews,Olivier Elemento,Daniel J. Nolan,Brendon M. Stiles,Shahin Rafii,Navneet Narula,Ramana V. Davuluri,Nasser K. Altorki,Vivek Mittal +17 more
TL;DR: Analysis of homogeneousStromal populations isolated directly from fresh clinical specimens can detect important stromal genes of therapeutic value and enhanced tumorigenic properties of lung cancer cells indicative of their potential as targets for therapy are demonstrated.
