Abstract: Oculopharyngeal muscular dystrophy (OPMD) is an adult-onset disease characterized by progressive eyelid drooping, swallowing difficulties and proximal limb weakness. The autosomal dominant form of the disease is caused by short (GCG)8–13 expansions in the PABP2 gene. This gene encodes the poly(A) binding protein 2 (PABP2), an abundant nuclear protein that binds with high affinity to nascent poly(A) tails, stimulating their extension and controlling their length. I nt his work we report that PABP2 is detected in filamentous nuclear inclusions, which are the pathological hallmark of OPMD. Using both immunoelectron microscopy and fluorescence confocal microscopy, the OPMD-specific nuclear inclusions appeared decorated by anti-PABP2 antibodies. In addition, the inclusions were labeled with antibodies directed against ubiquitin and the subunits of the proteasome and contained a form of PABP2 that was more resistant to salt extraction than the protein dispersed in the nucleoplasm. This suggests that the polyalanine expansions in PABP2 induce a misfolding and aggregation of the protein into insoluble inclusions, similarly to events in neurodegenerative diseases caused by CAG/polyglutamine expansions. No significant differences were observed in the steady-state poly(A) tail length in OPMD and normal myoblasts. However, the nuclear inclusions were shown to sequester poly(A) RNA. This raises the possibility that in OPMD the polyalanine expansions in the PABP2 protein may interfere with the cellular traffic of poly(A) RNA.

Abstract: We present the first hybrid measurement of the average muon number in air showers at ultrahigh energies, initiated by cosmic rays with zenith angles between 62° and 80°. The measurement is based on 174 hybrid events recorded simultaneously with the surface detector array and the fluorescence detector of the Pierre Auger Observatory. The muon number for each shower is derived by scaling a simulated reference profile of the lateral muon density distribution at the ground until it fits the data. A 1019eV shower with a zenith angle of 67°, which arrives at the surface detector array at an altitude of 1450 m above sea level, contains on average (2.68±0.04±0.48(sys))×107 muons with energies larger than 0.3 GeV. The logarithmic gain dlnNμ/dlnE of muons with increasing energy between 4×1018eV and 5×1019eV is measured to be (1.029±0.024±0.030(sys)).

Abstract: During the last decades, climate and land use changes led to an increased prevalence of megafires in Mediterranean-type climate regions (MCRs). Here, we argue that current wildfire management policies in MCRs are destined to fail. Focused on fire suppression, these policies largely ignore ongoing climate warming and landscape-scale buildup of fuels. The result is a "firefighting trap" that contributes to ongoing fuel accumulation precluding suppression under extreme fire weather, and resulting in more severe and larger fires. We believe that a "business as usual" approach to wildfire in MCRs will not solve the fire problem, and recommend that policy and expenditures be rebalanced between suppression and mitigation of the negative impacts of fire. This requires a paradigm shift: policy effectiveness should not be primarily measured as a function of area burned (as it usually is), but rather as a function of avoided socio-ecological damage and loss.

Abstract: miRNAs were recently implicated in the pathogenesis of numerous diseases, including neurological disorders such as Parkinson's disease (PD). miRNAs are abundant in the nervous system, essential for efficient brain function and play important roles in neuronal patterning and cell specification. To further investigate their involvement in the etiology of PD, we conducted miRNA expression profiling in peripheral blood mononuclear cells (PBMCs) of 19 patients and 13 controls using microarrays. We found 18 miRNAs differentially expressed, and pathway analysis of 662 predicted target genes of 11 of these miRNAs revealed an over-representation in pathways previously linked to PD as well as novel pathways. To narrow down the genes for further investigations, we undertook a parallel approach using chromatin immunoprecipitation-sequencing (ChIP-seq) analysis to uncover genome-wide interactions of α-synuclein, a molecule with a central role in both monogenic and idiopathic PD. Convergence of ChIP-seq and miRNomics data highlighted the glycosphingolipid biosynthesis and the ubiquitin proteasome system as key players in PD. We then tested the association of target genes belonging to these pathways with PD risk, and identified nine SNPs in USP37 consistently associated with PD susceptibility in three genome-wide association studies (GWAS) datasets (0.46≤OR≤0.63) and highly significant in the meta-dataset (3.36×10−4

Abstract: Among the multiple definitions for the Virtual Enterprise concept, the significance of two key elements: networking and cooperation, becomes clear. The lack of a unified definition is partially resulted by the large diversity in classes of networked organizations, which calls for the establishment of a taxonomy of virtual enterprises. An attempt to present some current definitions and a taxonomy is made. The first step in the analysis of virtual enterprises requires a careful study of various phases of its life cycle and characterization of the roles of various involved actors.
