Proteo-genomics of soluble TREM2 in cerebrospinal fluid provides novel insights and identifies novel modulators for Alzheimer disease
Li Wang,Niko Nykanen,Daniel Western,Priyanka Gorijala,Jigyasha Timsina,F. Li,Z.H. Wang,M Ali,C. Yang,Marta Marquié,Mercè Boada,Ignacio Alvarez,Mercè Miquel,Parviz Daniel Hejazi Pastor,Agustín Ruiz,Raquel Delgado Puerta,Adelina Orellana,Jarod Rutledge,Hamilton Oh,Michael D. Greicius,Yann Le Guen,R. Perrino,Tony Wyss-Coray,Ayanna Jefferson,T. Hohman,N. Graff-Radford,Hiroshi Mori,Alison Goate,Jeffrey Levin,Yun Ju Sung,Carlos Cruchaga +30 more
TL;DR: In this paper , the authors performed the largest genome-wide association study (GWAS) and identified four loci for CSF sTREM2 in 3350 individuals of European ancestry.
Journal ArticleDOI
PHOSPHOLIPASE D3 CONTRIBUTES TO ALZHEIMER’S DISEASE RISK VIA DISRUPTION OF Aβ CLEARANCE THROUGH THE LYSOSOME
Celeste M. Karch,Simon Hsu,Rita Martinez,Joanne Norton,John R. Cirrito,Jin-Moo Lee,Maria Cuervo,Carlos Cruchaga,Alison Goate +8 more
TL;DR: PLD3 may impact amyloid accumulation and AD risk through disrupted microglia function as PLD3 is enriched in disease 59 associatedMicroglia in human brains and these findings demonstrate that PLD 3 regulates.
Journal ArticleDOI
Progressive white matter injury in autosomal dominant Alzheimer’s disease is strongly associated with cerebral microbleeds and neurodegeneration
Zahra Shirzadi,Stephanie A. Schultz,Wai-Ying W Yau,Nelly Joseph-Mathurin,Kejal Kantarci,Gregory M. Preboske,Clifford R. Jack,Martin R. Farlow,Anne M. Fagan,Jason Hassenstab,Mathias Jucker,John C. Benzinger,Hamid R. Sohrabi,Ralph N. Martins,L. Schultz,Randall J. Bateman,Keith A. Johnson,Reisa A. Sperling,Steven M. Greenberg,Jasmeer P. Chhatwal +43 more
TL;DR: In this article , the white matter (WM) injury visible on MRI is a common finding in Alzheimer's disease (AD) and is often attributed to small vessel ischemic changes secondary to increased systemic vascular risk.
Journal ArticleDOI
Validation of newly derived polygenic risk scores for dementia in a prospective study of older individuals.
Chenglong Yu,Joanne Ryan,Suzanne G Orchard,Catherine Robb,Robyn L. Woods,Rory Wolfe,Alan E. Renton,Alison Goate,Amy Brodtmann,Raj C. Shah,Trevor T.-J. Chong,Kerry M. Sheets,Christopher Kyndt,Ajay Sood,Elsdon Storey,Anne M. Murray,John J McNeil,Paul Lacaze +17 more
TL;DR: This paper used Cox models and area under the curve (AUC) to validate new polygenic risk scores (PRS-83SNP, SBayesR, and CS) compared with an older PRS-23SNP in 12,031 initially healthy participants ≥70 years of age.
Journal ArticleDOI
Reply to Bertram et al.
Andrew Grupe,Yonghong Li,Charles M. Rowland,Tony Hinrichs,Peter Holmans,John Hardy,Michael Conlon O'Donovan,Michael John Owen,Julie Williams,Alison Goate +9 more
TL;DR: The study by Bertram and colleagues failed to replicate, in two family-based sample sets, the association of rs498055 with Alzheimer disease (AD [MIM 104300]) that was observed in four large, well-characterized case-control sample sets; there are several differences between the studies that may have contributed to these discrepant findings.
