TL;DR: Parkinsons disease (PD) is genetically associated with the H1 haplotype of the MAPT 17q.21.31 locus, although the causal gene and variants underlying this association have not been identified as mentioned in this paper.

TL;DR: In this paper, the authors applied a genetic-based approach to sample selection; their cohort included carriers of AD pathogenic mutations (APP, PSEN1), risk variants in TREM2, and the resilience variant (rs1582763) in the MS4A cluster associated with cerebrospinal fluid (CSF) soluble TREM 2 levels.
Journal ArticleDOI
Novel progranulin variants do not disrupt progranulin secretion and cleavage
TL;DR: Deep sequencing of families exhibiting late-onset dementia revealed several novel variants in progranulin gene (GRN) that behave like PGRN wild-type protein, suggesting that these variants represent rare polymorphisms, however, it remains possible that they affect other aspects of P GRN function or represent risk factors for dementia when combined with other modifying genes.
Journal ArticleDOI
Astrocyte-secreted glypican-4 drives APOE4-dependent tau hyperphosphorylation
TL;DR: It is reported that the astrocyte-secreted protein glypican-4 (GPC-4), which is identified as a binding partner of APOE4, drives tau hyperphosphorylation and further shows that APoe4-mediated surface trafficking of APoe receptor low-density lipoprotein receptor-related protein 1 through GPC- 4 can be a gateway to tau spreading.
